Abstract:
Objective: To detect the gene expression of the co-stimulatory molecular ICOS/ICOSL mRNA in gastric adenocarcinomas and homologous paraneoplastic tissue using the technology of real-time fluorescent quantitative polymerase chain reaction (real-time fluorescence PCR), in order to explore the role of co-stimulatory molecules ICOS/ICOSL in the tumorigenesis and development of gastric adenocarcinoma. Methods:Forty-two gastric adenocarcinoma patients (age ranging 39-85 years), confirmed by pathologic section and without anticancer treatments such as radio-chemotherapy, etc., were selected as the research participants. The cDNA of tissue samples was synthesized by RT-PCR after extracting the total RNA, then the target sequences of ICOS/ICOSL cDNA were amplified by Taqman probe real-time fluorescence PCR using the template of tissue cDNA. The expression of ICOS/ICOSL mRNA of the gastricadenocarcinoma tissue and the adjacent tissue was quantitatively measured.Results: In the 42gastric carcinoma tissue samples, the relative mean expression of ICOS mRNA was higher than the adjacent tissue (P=0.005 ), but the difference of ICOSL mRNA between them had no statistical significance ( P>0.05).The expression of ICOSL mRNA of different sex, age, clinical staging and cellular differentiation of participants had no statistical significance ( P>0.05). Conclusion: The expression level of ICOS mRNA in gastric adenocarcinoma tissue was sharply reduced compared with their adjacent tissue and this may cause the immune system evasion of carcinomas.
