人食管癌细胞缺氧培养条件下HIF-1α和VEGF表达的研究*

Expression of HIF-1α and VEGF in Human Esophageal Carcinoma Cells

  • 摘要: 探讨人低分化食管鳞癌细胞系EC- 109、CaEs-17在缺氧条件下,缺氧诱导因子-1 α 及血管内皮生长因子随缺氧时间不同,在基因水平和蛋白水平上的表达。方法:1)体外培养人低分化食管鳞癌细胞系EC- 109、CaEs-17,利用免疫荧光染色方法观察细胞在缺氧培养条件下HIF-1 α 和VEGF蛋白表达情况。2)提取总RNA,逆转录为cDNA ,并进行实时PCR ,观察细胞在不同缺氧时间,hif- 1 α 及vegf 基因表达峰度及相互关系。结果:1)EC- 109、CaEs-17细胞在缺氧培养条件下HIF-1 α 、VEGF蛋白表达增加。2)EC- 109、CaEs-17细胞在缺氧培养条件下hif- 1 α 、vegf 基因表达上调。3)EC- 109、CaEs-17细胞在缺氧条件下vegf 表达峰度上调滞后于hif- 1 α 表达,自缺氧培养8h 起,hif- 1 α 基因表达峰度持续高于vegf 基因表达。结论:缺氧环境是诱导人食管癌细胞缺氧诱导因子-1α和血管内皮生长因子表达上调的因素之一,vegf 基因表达滞后于hif- 1α基因表达。

     

    Abstract: Objective: To investigate the expression of hypoxia inducible factor- 1 α (HIF-1 α) and vascular endothel ial growth factor (VEGF) at the mRNA and protein levels in esophageal squamous cell carcinoma (ESCC) cell lines EC-109 and CaEs- 17at different time points under hypoxic conditions. Methods:Human ESCC cell lines EC-109 and CaEs- 17 were cultured in vitro and the expression of HIF- 1 α and VEGF proteins under hypoxic condi tions was detected wi th immu-nofluorescence. Real-time PCR was used to detect HIF-1 α and VEGF mRNA expression. Results: The mRNA and protein expression levels of HIF-1 α and VEGF in EC-109 and CaEs- 17cells were increased under hypoxic conditions. The in -crease in VEGF mRNA and protein expression appeared later than that of HIF-1 α under hypoxic condi tions, and HIF-1 α mRNA and protein expression was higher than VEGF mRNA and protein expression at8 hours after induction of hypoxia. Conclusion: Hypoxic conditions can upregulate the mRNA and protein expression of HIF-1 α and VEGF in ESCC cel ls. The increase in VEGF expression appears later than that of HIF- 1α.
