  • 摘要: 四跨膜蛋白是一组具有四个高度疏水的跨膜结构域的低分子量细胞膜表面蛋白,可广泛参与细胞生长、粘附、迁移等多种生命活动。四跨膜蛋白超家族(TM4SF)可与包括四跨膜蛋白、整合素、生长因子及其受体、HLA家族、主要组织相容复合物(MHC)等在内的多种细胞表面分子相连接,形成以TM4SF为核心的四跨膜蛋白网络。四跨膜蛋白网络可通过直接或间接作用于细胞信号通道从而影响肿瘤细胞的粘附、分化、迁移及侵袭。目前研究表明四跨膜蛋白CD151、CD82、Tspan8及Tspan1与肝癌密切相关,可参与调控肝癌细胞的粘附、增殖、分化、迁移及侵袭等过程来影响肝癌的发生发展。KAI1/CD82为肝癌的抑制基因,上调其表达可以抑制肝癌细胞的迁移和侵袭;CD151在肝癌组织中大量表达,提示CD151或联合CD151/c-Met是可能预测肝癌的进展新指标,并且CD151可能用于肝癌的靶向治疗;而Tspan8(CO-029)及Tspan1(NET-1)则可用于预测肝癌的进展及预后。

     

    Abstract: Tetraspanins are a family of small cell surface proteins characterized by four highly conserved transmembrane domains, which are thought to be involved in the regulation of a broad range of functions, including cell growth, conglutination, and migration. The tetraspanin superfamily ( TM4SF ) may be associated with other cell surface proteins such as tetraspanins, integrin growth factor and its receptor, the HLA family, and MHCs. They form the tetraspanin web wherein tetraspanins form the cores. The tetraspanin web indirectly or directly affects conglutination, differentiation, migration, and metastasis of the cancer cell by signal pathway. Current research studies show that CD151, CD82, Tspan8, and Tspan1 are closely associated with liver cancer, which may affect the progress of liver cancer by regulating conglutination, migration, and metastasis of liver cancer cells. KAI1/CD82 is a tumor suppressor gene that inhibits the migration and metastasis of liver cancer cell by upregulating the level of its expression. CD151 is expressed widely in liver cancer, which shows the possible use of CD151 overexpression and c-Met overexpression in predicting the progression of liver cancer, and CD151 may be potential molecular therapeutic targets for liver cancer. The progression and the prognosis of liver cancer can be predicted using Tspan8 ( CO-029 ) and Tspan1 ( NET-1 ).
