黄芩素对人肝癌细胞株SMMC-7721体外迁移及侵袭的影响

Effect of Baicalein on the In Vitro Migration and Invasion of Human Hepatocellular Carcinoma Cell Line SMMC-7721

      目的  通过观察黄芩素对人肝癌细胞株SMMC-7721体外迁移侵袭能力及Ezrin、MMP-9、VEGF表达的影响, 探讨黄芩素对人肝癌细胞株SMMC- 7721体外迁移侵袭的作用及其可能机制。
      方法  体外培养SMMC-7721, 经黄芩素干预后用划痕愈合实验和侵袭小室实验分别检测黄芩素对SMMC-7721细胞迁移运动能力和侵袭能力的影响; RT-PCR和免疫细胞化学法检测黄芩素作用于SMMC-7721细胞48h后对Ezrin、MMP-9、VEGF mRNA和蛋白表达的影响。
      结果  划痕愈合实验显示各浓度实验组划痕愈合宽度占原宽度的比例比对照组明显下降(P < 0.05); 体外侵袭实验显示实验组穿过人工基底膜胶的细胞数明显减少(P < 0.05); 实验组Ezrin、MMP-9、VEGF mRNA和蛋白表达均明显下调(P < 0.05)。
      结论  黄芩素可能通过直接或间接下调Ezrin、MMP-9、VEGF mRNA和蛋白的表达而抑制人肝癌细胞株SMMC- 7721体外迁移侵袭, 传统中药黄芩可望为肝癌临床治疗提供新的安全、有效、价廉的治疗方法。

     

    Abstract:
      Objective  This study aims to investigate the possible inhibitory effect of baicalein on the migration and invasion of the human hepatocellular carcinoma cell line SMMC-7721 by studying the expression of ezrin, matrix metalloproteinase-9 (MMP-9), and vascular endothelial growth factor (VEGF) in the said cells.
      Methods  Wound-healing and transwell chamber assays were performed to investigate the migration and invasion of baicalein-treated SMMC-7721 cells. The mRNA and protein expression of ezrin, MMP-9, and VEGF in baicalein-treated SMMC-7721 cells were determined using reverse transcription-polymerase chain reaction and immunocytochemistry, respectively.
      Results  The results of the wound-healing assay showed that the percentage of wound closure in the experimental group declined (P < 0.05). Meanwhile, the results of the transwell assay showed that the number of SMMC-7721 cells permeating through the artificant membrane in the baicalein-treated group also decreased (P < 0.05). The protein and mRNA expression levels of ezrin, VEGF, and MMP-9 were downregulated (P < 0.05).
      Conclusion  Baicalein may partly induce anti-neoplastic effectiveness by directly or indirectly inhibiting the expression of ezrin, VEGF, and MMP-9. This Baical Skullcap root extract can be used in clinical applications and may be used as a new, safe, and effective plant medicine for hepatocellular carcinoma.
