Abstract:
Objective To investigate the effect of anti-MUC 1 monoclonal antibody (Mab) C595 on the growth and apoptosis of human ovarian cancer cell line OVCAR-3.
Methods The expression of MUC 1 on OVCAR-3 cells was examined by immunofluorescence labeling and flow cytometry using MabC595. The cell proliferation was measured by MTT assay and colony formation assay. TUNEL method was used to detect apoptosis. Cytochrome c and Caspase-3 activity were measured by ELISA.
Results Mab C595 inhibited the growth of OVCAR-3 cells in a concentration-and time-dependent manner. TUNEL showed that Mab C595 induced apoptosis of OVCAR-3. ELISA analysis showed that cell death following treatment was associated with the release of cytochrome c and Caspase-3 concentrations.
Conclusion Mab C595 inhibits the proliferation and induces cell apoptosis in human ovarian cancer cell line OVCAR-3. The mechanisms of cell apoptosis induced by Mab C595 may be through upregulating Caspase-3 activity.
