Relationship between Soxll gene Methylation and Gastric Cardia Adenocarcinoma

      目的  检测贲门腺癌(gastric cardia adenocarcinoma, GCA)组织中Wnt通路相关因子Sox17基因的甲基化状态及mRNA表达情况, 探讨其与贲门腺癌发生的关系。
      方法  应用甲基化特异性PCR(methylation specific PCR, MSP)及RT-PCR的方法检测126例贲门腺癌及相应癌旁非肿瘤组织中Sox17基因的甲基化状态及mRNA表达情况。
      结果  在126例贲门腺癌组织中, Sox17基因的甲基率为86.5%(109/126), 明显高于癌旁非肿瘤组织(P < 0.001);癌组织中Sox17基因的甲基化率与贲门腺癌的TNM分期及淋巴结转移均无关; 癌组织中该基因mRNA的阳性表达率为58.7%(74/126), 明显低于癌旁非肿瘤组织(P < 0.001)。其mRNA阳性表达率与该基因的甲基化状态呈明显负相关(P < 0.01)。
      结论  Sox17基因在贲门腺癌中呈高甲基化状态, 可作为Wnt拮抗基因甲基化谱中的一个新成员。

     

    Abstract:
      Objective  To investigate the state of methylation of the Soxl7 gene in gastric cardia adenocarcinoma(GCA).
      Methods  Methylation-specific polymerase chain reaction(MSP) and real-time polymerase chain reaction(RT - PCR) methods were performed to examine the CpG methylation of the Sox 17 gene and its mRNA expression in 126 GCA samples and in the corresponding paraneoplastic tissues.
      Results  The frequency of Sox17 gene methylation in the GCA tissues was 86.5%(109 / 126), which is significantly higher than that in the paraneoplastic tissues(P < 0.01).The hypermethylation of this gene showed no correlation with the clinical stage and pathologic classification of cancer tissues(P > 0.05).The positive rate of Sox17 mRNA expression in the GCA tissues was 58.7%(74 / 126), which is lower than that in the paraneoplastic tissues(P < 0.01).The mRNA expression correlated with the frequency of Soxl7 methylation(P < 0.01).
      Conclusion  Soxl7 methylation is a frequent event in GCA.Therefore, this gene can be included in the list of DNA-hypermethylated Wnt antagonist genes in GCA.
