Expression and Relationship of Notch 1, Jagged1 and COX-2 in Gastric Cancer

      目的  研究Notch1、Jagged1和COX-2在胃癌组织中的表达情况, 分析其与临床病理特征及Hp感染的关系。
      方法  应用免疫组织化学S-P法, 采用组织芯片技术, 检测60例胃癌组织、60例癌旁组织及20例正常胃黏膜组织中Notch1、Jagged1和COX-2蛋白的表达; 采用Warthin-starry染色法检测各组织中Hp感染状况。
      结果  Notch1与Jagged1在胃癌组中阳性表达率(40.0%, 70.0%)低于癌旁组(81.7%, 96.7%)和正常组(85.0%, 100.0%), 差异有统计学意义(P < 0.05)COX-2在胃癌组中的阳性表达率(60.0%)高于癌旁组(35.0%)和正常组(0), 差异有统计学意义(P < 0.05)。Notch1和Jagged1与胃癌的临床病理特征均无关(P > 0.05)。COX-2的表达与胃癌的分化程度、浸润深度(P=0.013, P=0.013)有关。Notch1与Jagged1在胃癌组中的表达呈正相关(r=0.460, P < 0.005);Notch1与COX-2表达呈负相关(r=-0.314, P > 0.05);Jagged1与COX-2表达无相关性(r=-0.175, P > 0.05)。Hp在胃癌组中阳性感染率低于癌旁组(76.6%, 91.7%), 差异有统计学意义(P < 0.05)。Notch1、Jagged1和COX-2在胃癌组中Hp感染阳性与Hp感染阴性的阳性表达率相比, 差异无统计学意义(均P > 0.05)。胃癌组织中Notch1、Jagged1和COX-2的表达与Hp感染率无关(P > 0.05)。
      结论  胃癌中存在Notch信号通路的异常, Notch1和Jagged1的低表达可能导致COX-2的高表达, 最终导致胃癌的发生发展; Hp感染可能在早期胃癌的发生中起重要作用。

     

    Abstract:
      Objective  To investigate the expression ofNotchl, Jaggedl and COX-2 in gastric cancer as well as their correlations with the clinical pathology and Hp infection.
      Methods  The expression of Notchl, Jaggedl and COX-2 proteins were detected in 60 cases of gastric cancer, 60 cases of normal gastric mucosa tissue, and 20 cases of normal gastric tissue by tissue micmarray technique and immunohistochemical staining. For detection of Hp infection, Warthin-starry staining was used.
      Results  The positive expression rates of Notchl and Jaggedl in gastric cancer tissue (40.0%, 70.0%) were lower than that in normal gastric mucosa tissue (81.7%, 96.7%) and normal gastric tissue (85.0%, 100%), there was statistical significance (P < 0.05).The positive expression rate of COX-2 in gastric cancer tissue (60.0 %) was higher than that in normal gastric mucosa, normal gastric tissue (35.0%, 0.0%), there was statistical significance (P < 0.05).The expression of Notchl and Jaggedl were not correlated with Clinic Pathologic Feature (P > 0.05). The expression of COX-2 was closely correlated with the pathologic differentiation degree and depth of invasion (P= 0.013, P= 0.013). The expression of Notchl was correlated with Jaggedl (r= 0.460, P < 0.005);The expression of Jaggedl was no relationship with COX-2 (r= -0.175, P < 0.05); The level of Notchl expression was negatively correlated with COX-2 (r= -0.314, P > 0.05).The infection rate of lip in gastric cancer (76.6 %) was lower than that in normal gastric mucosa tissue (91.7%), there was statistical significance (P < 0.05). The expression of Notchl, Jaggedl and COX-2 were no correlation with the infection of lip (P > 0.05).
      Conclusion  Aberrant signaling within the Notch pathway in gastric cancer, down-expression of Noteh l and Jagged l induced COX-2 over-expression, finally gastric tumor forming. Hp infection may be plays an important role at the early stage of development of gastric cancer.
