Abstract:
Objective This study was designed to investigate the mechanism and effect of LBH589, a novel histone deacetylase inhibitor, on the growth and apoptosis of the human ovarian cancer cell line OVCAR-3.
Methods The cells were treated with LBH589 a different concentrations.Cell proliferation was determined using the methyl thiazol tetrazolium assay.The apoptotic rate of the cells was detected by AO/EB double-staining.Protein expressions of polyADP-ribose polymerase(PARP), caspase-3, bel-2, and bax were analyzed by western blot assay.
Results LBH589 significantly inhibited the proliferation of OVCAR-3 cells at a concentration of 0.12 μM/L based on a 48 h half-inhibitory concentration(IC50).Western blot assay showed that the expressions of cleaved PARP-85KD, caspase-3, and bax were increased, but bcl-2 expression was downregulated.
Conclusion LBH589 in vitro can significantly inhibit the proliferation and induce apoptosis of the human ovarian cancer cell line OVCAR-3.
