FISH技术在淋巴瘤分子特征中的研究和应用进展

Progress in the application of FISH for the molecular characterization of lymphomas

  • 摘要: 荧光原位杂交(FISH)技术是一种用荧光素标记的核酸探针和特定片段杂交,通过荧光信号变化对染色体或基因异常进行定性、定位及相对定量分析的方法。淋巴瘤作为常见的恶性肿瘤之一,其分类复杂,不同种类的淋巴瘤常涉及某些特定基因和染色体的改变,并且不同分子特征的淋巴瘤其预后差异也较大。弥漫性大B细胞淋巴瘤(DLBCL)、黏膜相关淋巴组织(MALT)淋巴瘤、滤泡淋巴瘤(FL)、间变性大细胞淋巴瘤(ALCL)等均具有特征性的染色体和基因异常,且不同分子亚型的淋巴瘤也涉及不同的染色体异常。目前FISH技术已被广泛应用于淋巴瘤的分子遗传学研究,并为临床诊断和预后监测提供了较为客观科学的依据。本文对FISH技术在不同类型淋巴瘤分子特征的研究和应用进行综述。

     

    Abstract: Fluorescence in situ hybridization (FISH) is a cytogenetic technique that detects changes in target genes and chromosomes using the hybridization of fluorescent-labeled probes to specific DNA fragments on chromosomes. Lymphomas are among the most common malignant tumors worldwide. These tumors have complex subtypes with various responses to therapy and different prognosis. The different lymphomas demonstrate abnormal gene and chromosome characteristics, which may be associated with patient prognosis, such as in diffuse large B cell lymphomas, mucosa-associated lymphoid tissue lymphomas, follicular lymphomas, and anaplastic large cell lymphomas. FISH has been widely used to study the molecular genetics of lymphomas and to provide scientific evidence and clues for clinical diagnosis and prognosis. This article gives a brief review of the applications of FISH in the study and diagnosis of different types of lymphomas.
