参芪扶正注射液对癌因性疲乏中肾上腺皮质作用的研究

Therapeutic mechanism of shen qi fu zheng zhu she ye toward the adrenal cortex ultrastructure in cancer-related fatigue

      目的  研究肾上腺皮质在癌因性疲乏发病机制中的作用以及参芪扶正注射液的治疗作用。
      方法  1) 癌因性疲乏动物模型的建立:BALB/C裸鼠130只, 把裸鼠随机分为正常对照组10只、模型对照组 (30只, 其中轻、中、重度疲乏组各10只) 、治疗组 (90只, 其中轻、中、重度疲乏组各30只) (每组再分3个治疗小组分别是培本扶正治疗组、化疗组、培本扶正联合化疗组, 每组各10只) ;以人胃癌BGC-823细胞株, 取2.5×106/mL细胞浓度, 0.2 mL/只裸鼠注射造原位移植癌;应用5-氟尿嘧啶及参芪扶正注射液单独和联合注射做治疗对照;应用水迷宫实验、旷场实验、鼠尾悬挂实验、力竭游泳实验对原位移植癌裸鼠进行行为学检测及癌因性疲乏状态的判定。2) 透射电镜观察裸鼠肾上腺皮质束状带超微结构。3) 高效液相色谱技术测定裸鼠外周血皮质醇含量。
      结果  轻度、中度和重度疲乏状态的各治疗组的裸鼠中, 血浆皮质醇的含量均低于正常对照组的裸鼠, 且差异有统计学意义。肾上腺皮质细胞核和线粒体的改变从正常对照到轻度、中度和重度疲乏组呈现一种衰老凋亡的表现。
      结论  肾上腺皮质的功能状态在癌因性疲乏的发病机制中起着一定的作用, 而参芪扶正注射液则通过改善肾上腺皮质的功能状态而对癌因性疲乏具有一定的治疗缓解作用。

     

    Abstract:
      Objective  The present study aimed to examine the therapeutic mechanism of shen qi fu zheng zhu she ye toward the adrenal cortex ultrastructure in cancer-related fatigue (CRF) .
      Methods  BALB/C nude mice (n=130) were used to establish animal models for the CRF.The nude mice were categorized into control (n=10) , model (n=30) , and treatment (n=90) groups.We injected 0.2 ml of the cell culture (cell density of human stomach cancer BGC-823 cell line = 2.5×106/mL) into the nude mice.During cancer transplantation, 5-fluorine uracil and shen qi fu zheng zhu she ye were used in making treatment comparisons of the injected and untreated mice.Water maze, tail hanging, and swimming capability experiments, as well as open field test, were conducted to determine the behavior of the animal models.The ultrastructure of the adrenal cortex was observed by an electron microscope.High-performance liquid chromatography was employed to determine the cortisol levels.
      Results  The cortisol levels in the mild, moderate, and severe fatigue groups were significantly lower than the control group.The change in the adrenal cortex ultrastructure was nonsignificant.
      Conclusion  The adrenal cortex ultrastructure plays a role in the CRF, especially when the body was tired.Shen qi fu zheng zhu she ye exhibited therapeutic action toward the adrenal cortex ultrastructure in CRF.
