Abstract:
Objective This work aimed to investigate the effect of Pseudomonas Aeruginosa Injection (PA-MSHA) on the growth of human carcinoma cell lines (hepatocellular carcinoma BEL-7402 cell, breast carcinoma MDA-MB-231 cell, gastric carcinoma MKN-45 cell, lung carcinoma A549 cell and colorectal carcinoma SW620) in vitro.The comparison and analysis of the PA-MSHA sensitivity was performed.
Methods The five human carcinoma cell lines were treated with Pseudomonas Aeruginosa Injection (PA-MSHA 1.8×109/mL) of gradient dilution for 72 h in vitro respectively, and observed by optical microscope.Methyl thiazolyl tetrazolium (MTT) was used to analyze cell proliferation, and linear fitting method to calculate IC50.
Results We found that PA-MSHA inhibited the growth of all of the carcinoma cells.The result of MTT showed dose-dependent inhibition effect of PA-MSHA on proliferation of carcinoma cells in vitro.IC50 of PA-MSHA was 2.12×108/ml (BEL-7402), 1.95×108/ml (MDA-MB-231), 2.09×108/ml (MKN-45), 1.59×108/ml (A549) and 1.32×108/ml (SW620), respectively.
Conclusion Pseudomonas Aeruginosa Injection directly inhibited proliferation of human carcinoma cells in vitro, and had inhibitory effect of broad spectrum on different tumor cell sources.Drug sensitivity of PA-MSHA in five carcinoma cell lines was SW620 > A549 > MDA-MB-231 > MKN-45 > BEL-7402.
