Role of HIF-1α in hypoxic microenvironment of tumor and the targeted therapy

  • 摘要: 乏氧现象广泛存在于实体肿瘤中。乏氧诱导因子-1α (hypoxia inducible factor 1α, HIF-1α) 通过参与多种靶基因的转录调控影响肿瘤细胞的能量代谢、增殖和凋亡, 使细胞及组织产生一系列反应以适应缺氧环境, 同时促进肿瘤血管形成, 也增加肿瘤自身的侵袭性及对放化疗的抵抗性。以HIF-1α为靶点的治疗将可能成为肿瘤治疗的重要手段。

     

    Abstract: Hypoxia is a common characteristic in solid tumors.With the regulation of the transcription of multiple target genes, HIF-1α plays an important role in the energy metabolism, proliferation, and apoptosis of tumor cells.HIF-1α contributes to the adaptation of tumor cells and tissue to hypoxia stress through these processes.It also promotes tumor angiogenesis and increases their invasive ability and the resistance to radio and chemotherapy.Cancer treatment targeting on HIF-1α is an important potential therapy.
