Aurora-A激酶的研究进展

Research development of Aurora-A kinase

  • 摘要: Aurora激酶家族是一组近年来发现的调节中心体和微管功能的丝氨酸/苏氨酸蛋白激酶,在细胞有丝分裂的正常进程中起着非常重要的作用。相关研究表明在一些实体肿瘤如乳腺癌、卵巢癌、食管癌、结肠癌、肺癌及膀胱癌等肿瘤中均出现Aurora-A基因的高表达。目前,Aurora-A激酶作为肿瘤治疗的一个新的靶点,已经逐渐引起人们重视,其小分子抑制剂的研究和使用,尤其是与化疗药物的联合应用,将成为今后恶性肿瘤治疗的新方向。本文对近几年国内外在Aurora-A激酶的研究,对其生物学功能、与肿瘤的关系及其抑制剂的研究进展进行综述。

     

    Abstract: Aurora-A is a mitotic serine/threonine kinase that is evolutionally conserved and localized at the centrosome. Aurora-A activation is required for mitotic entry, centrosome maturation, and centrosome separation. Aurora-A is frequently amplified and/or over-expressed in breast, ovarian, esophageal, colon, lung, and bladder cancers. Aurora-A has recently become a new target of malignant tumors. The Aurora-A inhibitor can be combined with other chemotherapeutic drugs to provide a new way for cancer treatment. In this study, we review the function and inhibitor of Aurora-A kinase.
