  • 摘要: 哺乳动物Hedgehog基因家族包括3个成员:Sonic hedgehog(SHH)、Desert hedgehog(DHH)和Indian hedgehog(IHH)。SHH信号通路主要由分泌型信号糖蛋白SHH配体、跨膜蛋白受体PTCH和另一跨膜蛋白SMO的复合物,以及3种GLI蛋白的下游转录因子GLI1、GLI2、GLI3组成。作为最经典的信号通路之一,SHH信号通路在胚胎发生、发育和组织更新等过程中发挥至关重要作用。GLI1是该通路中最重要转录因子之一,其异常活化与纤维化修复、肿瘤的发生发展与侵袭转移、肿瘤耐药和预后等密切相关。本文就GLI1与肿瘤发生与侵袭转移作用机制进行综述。
