Research progress on the relationship between mitochondrial DNA variation and tumorigenesis

摘要: 肿瘤的发生发展是一个较为复杂的生物学过程,其中线粒体在肿瘤细胞代谢过程中扮演重要角色。线粒体DNA(mitochondrial DNA,mtDNA)变异包括点突变、缺失、插入以及拷贝数变异等,这些变异可能参与肿瘤细胞的增殖、生长、侵袭和转移。本文就mtDNA突变、拷贝数改变、mtDNA与核基因(nuclear DNA,nDNA)的比值,以及mtDNA变异与肿瘤发生发展的相关性研究及临床进展进行综述,旨在为深入认识mtDNA变异与肿瘤发生发展的关系并为临床治疗提供更多的参考依据。Abstract: The occurrence and development of tumors is a biological process that is extremely complex. In depth research gradually revealed that mitochondria play an important role in the metabolism of tumor cells. The types of mitochondrial DNA (mtDNA) mutations identified contain point mutations, deletions, insertions, and copy-number variations. These mutations may be involved in the proliferation, growth, invasion, and metastasis of tumor cells. In this study, mtDNA mutations, copy-number changes, correlation of mtDNA with nuclear DNA, and the progress gained in the development of the relationship between mtDNA mutations and tumorigenesis are detailed. This review aims to provide additional references to improve the understanding of the relationship between mtDNA mutations and tumor progression.
