白藜芦醇对宫颈癌HeLa细胞表皮生长因子表达的影响

The Effect of Resveratrol on the Expression of Epidermal Growth Factor in HeLa Cervical Cancer Cells

  • 摘要: 目的 :通过体外实验,研究白藜芦醇对宫颈癌细胞表皮生长因子(Epidermal growth factor,EGF)表达的影响,探讨白藜芦醇对宫颈癌侵袭影响的作用机制。 方法 :采用Transwell侵袭系统,检测白藜芦醇对宫颈癌HeLa细胞侵袭能力的影响;扫描电子显微镜,观察白藜芦醇对肿瘤细胞伪足形态的影响;RT-PCR、Western blot法了解白藜芦醇对HeLa细胞EGF mRNA及蛋白表达的影响。 结果 :Transwell侵袭实验结果表明,白藜芦醇1mM、0.5mM两组平均穿过的细胞数分别为9.20±1.924,10.20±2.049与空白对照组及溶剂对照组(平均穿过的细胞数分别为39.80±2.387,39.00±3.808)相比,明显减少(P<0.05)。RT-PCR及Western blot结果表明,白藜芦醇能够明显降低宫颈癌细胞EGF mRNA及蛋白表达水平。同时,扫描电子显微镜观察到,经白藜芦醇作用后,HeLa细胞逐渐由多角形向圆形转变,周围生长出的片层状、不规则的伪足结构(包括板状伪足和丝状伪足)逐渐减少,伪足变短变细,且断裂增多。 结论 :白藜芦醇能够有效降低宫颈癌细胞EGF mRNA及蛋白的表达水平,进而对肿瘤细胞侵袭转移所必需的细胞装配—伪足结构的形成数量和质量产生影响。白藜芦醇通过此作用机制达到对宫颈癌细胞侵袭的抑制作用。

     

    Abstract: Objective : To investigate the effect of resveratrol on the expression of epidermal growth factor (EGF) and to identify the effect of resveratrol on the invasion of cervical cancer in vitro. Methods : The effect of resveratrol on invasive ability of HeLa cells was observed with a Transwell cell culture chamber. Changes in parapodium structure were observed with scanning electron microscopy. EGF mRNA and protein expression in HeLa cells was detected by RT-PCR and Western blot. Results : In the Transwell cell culture chambers, the average number of invading HeLa cells was 9.20±1.924 in the group with 1mM resveratrol and 10.20±2.049 in the group with 0.5mM resveratrol, significantly lower than those in the control group and the DMSO group (39.80± 2.387, 39.00±3.808, respectively) (P <0.05). The levels of EGF mRNA and protein were also decreased after treatment with resveratrol. The parapodium structures of cells cultured with resveratrol appeared to be broken down. Conclusion : Resveratrol can effectively decrease the expression of EGF and then destroy the parapodium structures of cervical cancer cells. Through this mechanism, resveratrol can inhibit the migration and invasion of cervical cancer cells in vitro.
