  • 摘要: 目的: MCL1(myeloid cell leukemia-1)与多种恶性肿瘤的发生、发展及转移相关。本文从临床样本和细胞水平检测胃癌组织中MCL1蛋白的表达水平,探讨MCL1蛋白在胃癌发生、发展中的作用。 方法: 采用S-P免疫组化法检测正常胃黏膜、癌旁胃黏膜和胃癌组织芯片中MCL1蛋白的表达,Western blot检测MCL1蛋白在人胃癌BGC823、SGC7901、MKN28细胞中的表达。 结果: 免疫组化结果显示,MCL1蛋白表达定位于细胞的胞浆,着色呈黄色至棕黄色;正常胃黏膜、癌旁胃黏膜、胃癌中MCL1蛋白阳性表达率分别为18.18%(4/22)、54.17%(26/48)、83.33%(75/90);MCL1蛋白在胃癌中的表达高于癌旁胃黏膜及正常胃粘膜,癌旁胃黏膜与正常胃粘膜MCL1蛋白表达亦具有明显差异(P<0.05);MCL1蛋白阳性表达率在伴淋巴结转移组为93.48%(43/46),无淋巴结转移组为72.72%(32/44),两者差异有显著性意义(P<0.05);而与性别、年龄和肿瘤大小无关。S-P免疫组化法与Western blot结果显示MCL1蛋白表达与胃癌分化程度呈负相关。 结论: MCL1蛋白表达与胃癌发生、分化程度和淋巴结转移相关。

     

    Abstract: Objective : To investigate the expression of MCL1 protein in gastric carcinoma and to explore the relation-ship between MCL1 protein expression and the development of gastric carcinoma. Methods : Expression ofMCL1 protein was examined immunohistochemically by the streptavidin-peroxidase conjugated method (SP)in normal mucosa, paracancerous tissues and gastric carcinoma. Western blot was used to detect the expres-sion of MCL1 protein in BGC823, SGC7901 and MKN28 gastric cancer cell lines. Results : MCL1 protein ex-pression was located in the cytoplasm. The positive rate of MCL1 protein expression was 18.18% (4/22) innormal gastric mucosa, 54.17% (26/48) in paracancerous tissues, and 83.33% (75/90) in gastric carcinomatissues. The positive rate of MCL1 protein expression in gastric carcinoma was significantly higher than that inparacancerous tissues ( P <0.05). The positive rate of MCL1 protein expression in tissues adjacent to carcino-ma lesions was significantly higher than that in normal gastric mucosa ( P <0.05). MCL1 expression rate was93.48% (43/46) in tissues from carcinoma patients with lymph node metastases and 72.72% (32/44) in thosewithout lymph node metastases, with a significant difference ( P <0.05). No correlation was found betweenMCL1 protein expression and gender or age. The expression of MCL1 was negatively correlated with the dif-ferentiation of gastric carcinoma. Conclusion : MCL1 protein expression is related to the carcinogenesis, differ-entiation and lymph node metastasis of gastric carcinoma.
