黄芪多糖与粒细胞集落刺激因子对化疗后骨髓功能的影响

The Effects of APS and G-CSF on Bone Marrow Function after Chemotherapy

  • 摘要: 目的 :了解化疗药物环磷酰胺(CTX)对骨髓粒细胞释放过程的影响,并比较黄芪多糖(APS)和粒细胞集落刺激因子(G-CSF)在连续给药后的作用特点。 方法 :利用免疫组织化学和ELISA方法分别检测药物干预后不同时间点小鼠骨髓粒细胞L-选择素(CD62L)表达情况及血清可溶性L-选择素(sCD62L)的浓度,同时计数外周血白细胞数量,并做统计学分析。 结果 :1)实验小鼠在给予化疗药物后外周血白细胞数量在第7、10、14天与化疗前(第0天)比较均出现显著降低(2.02±0.27、2.96±0.65、3.43±1.37比6.13±0.70,P均<0.05),但呈逐渐上升趋势。骨髓粒细胞CD62L阴性表达率第7天与化疗前比较无明显变化(13.20±1.30比14.25±1.89,P>0.05),第10天和第14天比化疗前均显著升高(25.00±1.41、25.00±2.65比14.25±1.89,P<0.05),且与外周血白细胞数量呈正相关(r=0.8988,P>0.05);2)APS和G-CSF均能提高小鼠经化疗药物干预后外周血白细胞数量,APS能在第7天快速下调骨髓粒细胞CD62L的表达;3)APS组血清sCD62L浓度波动范围为1.02~1.49μg/m(l任一时间点与化疗前比较P均<0.05);而G-CSF组第14天出现明显升高,与APS组比较有差异(2.04±0.33比1.49±0.17,P<0.05),且与另两个指标均具有正相关性。 结论 :实验小鼠在注射化疗药物第7天出现骨髓粒细胞释放功能障碍;APS可通过促进骨髓粒细胞释放来提高外周血白细胞数量,作用缓和、持久;而G-CSF主要促进骨髓粒细胞增殖,作用强烈,并可能促进幼稚粒细胞释放。

     

    Abstract: Objective : To investigate the effect of CTX on the ability of bone marrow to release granulocytes and to compare the efficacy of APS and G-CSF. Methods : We detected the expression of CD62L on granulocytes in bone marrow, the serum sCD62L concentration and the peripheral blood white blood cell count in 90 mice at different time points of chemotherapy. All data were statistically analyzed. Results : (1) White blood cell count was reduced on day 7, 10 and 14 after chemotherapy compared with that on day 0. No changes were found in the negative rate of CD62L on granulocytes in bone marrow on day 7 but elevation was observed on day10 and 14. (2) White blood cell count was increased in APS and G-CSF-treated groups after chemotherapy.The negative rate of CD62L on granulocytes in bone marrow was increased simultaneously in the group treat-ed with APS. (3) The range of serum sCD62L concentration was 1.02-1.49μg/ml in the APS group. The con-centration of sCD62L was increased on day 14 in the G-CSF group compared with the APS group. Conclu-sion : The granulocyte releasing function of the bone marrow was impaired on day 7 of chemotherapy in experi-mental mice. APS promotes granulocyte releasing and can help increase white blood cell count in the periph-eral blood, an effect that is mild and sustained. G-CSF promotes the proliferation of granulocytes and the re-lease of immature granulocytes.
