摘要: 目的: 探讨中药消癌平和榄香烯注射液抑制宫颈癌细胞HeLa增殖作用及其作用机制。 方法: 取对数生长期的HeLa细胞接种于96孔培养板(细胞数为3×10⁴/孔,100μl/孔),加入不同浓度的消癌平、榄香烯注射液及其混合液,收集不同处理组作用72h的HeLa细胞,通过细胞增殖抑制试验测定吸收度A值。 结果: 消癌平和榄香烯25～400μg/ml作用72h对HeLa细胞体外增殖有显著的抑制作用,呈剂量依赖性。消癌平及榄香烯各100μg/ml联合作用72h对HeLa细胞体外增殖有显著的抑制作用,呈剂量依赖性,且明显高于榄香烯或消癌平单药作用。消癌平和榄香烯在抑制肿瘤细胞增殖时均伴随有Bcl-2蛋白水平表达的下调。 结论: 中药消癌平和榄香烯注射液联合应用抑制HeLa增殖作用优于单药,其机理可能与Bcl-2基因下调有关。

Abstract: Objective: : To study the inhibitory function and mechanism of Xiaoaiping injection and Elemene injectionon cervix cancer HeLa cells in vitro. Methods : HeLa cells were treated with Xiaoaiping and Elemene at different concentrations. MTT assay, flow cytometry and morphological examination were applied to evaluate cellproliferation and the expression of Bcl-2 protein. Results : Xiaoaiping combined with Elemene could remarkably inhibit HeLa cells and decrease the expression of Bcl-2 protein. The cells were mainly arrested in G1stage and were in a differentiating tendency. Conclusion : Xiaoaiping combined with Elemene could inhibitHeLa cells in G1 stage and down-regulate the level of Bcl-2.
