肿瘤细胞悬液注射法制作兔食管癌移植瘤模型

Establishment of a VX2 Esophageal Carcinoma Model in Rabbits

  • 摘要: 目的: 利用肿瘤细胞悬液注射法建立稳定的兔食管癌移植瘤模型,观察其钡餐、MRI及病理表现。 方法: 冻存的VX2肿瘤细胞经兔体内接种成瘤后制成细胞悬液,手术暴露食管,直视下将肿瘤细胞悬液接种于兔食管壁,制作兔食管癌移植瘤模型,2周后经钡餐和MRI检测其影像学表现,处死兔子取标本行病理学检测。 结果: 食管内注射肿瘤细胞悬液2周后,兔子出现吞咽困难的症状,食管钡餐显示明显的充盈缺损,缺损内径平均约0.518cm;MRI显示兔食管壁内孤立性结节状肿瘤,病灶在T1WI上呈略低信号或等信号,在T2W I上呈高信号;瘤体和淋巴结标本病理切片镜下观察,证实成瘤及出现淋巴结转移;模型建立成功率为100%。 结论: 使用肿瘤细胞悬液注射法可成功建立兔VX2食管癌动物模型。

     

    Abstract: Objective: : To establish a stable model of VX2 esophageal carcinoma in rabbits by injecting a tumor cellsuspension and to characterize the tumor model by barium meal study, MRI and histological methods. Methods : A VX2 tumor cell suspension was injected into rabbits and then tumor nodules were removed and pulverized to make a VX2 tumor cell suspension. After surgically exposing the subject rabbit's esophagus, this VX2tumor cell suspension was injected into the wall of the esophagus. Two weeks later, MRI and barium mealstudies were performed to identify the imaging features of this tumor model. Rabbits were sacrificed and tumor specimens were fixed with formalin for pathological examination. Results : At two weeks after injecting thetumor cell suspension into the rabbit esophagus, symptoms of dysphagia appeared. Filling defects were observed in the esophagus during the barium meal study, with an average diameter of about 0.518 cm. Isolatedtumor nodules were detected by MRI, and the tumors were hypointense on T1-weighted images and hyperintense on T2-weighted images. The success rate of model establishment was 100%. Conclusion : A rabbit model for VX2 esophageal carcinoma has been established successfully by injecting tumor cell suspension.
