S100A9 Galectin7在同时放化疗敏感性不同宫颈癌中的表达及意义

The Expression of S100A9 and Galectin7 in Cervical Carcinoma Tissues with Different Concurrent Chemoradiotherapy Sensitivity

  • 摘要: 目的: 观察S100A9和Galectin7在同时放化疗敏感性不同宫颈癌组织中的表达及与宫颈癌同时放化疗敏感性的关系。 方法: 收集进行治疗前中晚期宫颈癌组织标本,病理诊断均为中分化鳞癌,放化疗后根据WHO实体瘤疗效判断标准,将收集的宫颈癌组织标本分为高敏感组和低敏感组。提取组织总蛋白,进行双向凝胶电泳和质谱分析鉴定差异表达蛋白,分别应用Westernblot和免疫组织化学方法检测组织中蛋白的表达。 结果: 筛选得到在同时放化疗敏感性不同宫颈癌组织中差异表达蛋白S100A9和Galectin7,它们在高敏感组中的表达均高于低敏感组。WesternBlot检测结果与蛋白质组学结果一致。免疫组化结果显示S100A9在高敏感组中的表达强度和表达率(88.3%,53/60)均显著高于低敏感组(28.6%,10/35),Galectin7在高敏感组中的表达强度及表达率(91.7%,55/60)显著高于低敏感组(40%,14/35),差异均有统计学意义。 结论: S100A9和Galectin7的表达与宫颈癌同时放化疗敏感性有关,可能作为同时放化疗敏感性预测的标志物。

     

    Abstract: Objective: To observe the expression of S100A9 and Galectin7 in cervical carcinoma tissues with differ-ent concurrent chemoradiotherapy sensitivity and to analyze its significance. Methods: The fresh carcinoma tissues werecollected from 10 untreated cervical carcinoma patients. According to the response to concurrent chemoradiotherapy, thetissues were classified into two groups: the high sensitivity group(HS) and the low sensitivity group(LS). Protein separationwas performed by two-dimensional gel electrophoresis (2-DE). The peptide mass fingerprintings (PMF) were acquired bymatrix assisted laser desorption/ionization time-of-flight mass spectrometry(MALDI-TOF-MS) and the proteins were iden-tified by data searching in the Mascot-database. The differentially expressed proteins were assayed by Western blot andimmunohistochemistry. Results: Most of the gels were clear and successfully and reproductively analyzed. It showed thatthe expression of S100A9 and Galectin7 was higher in the HS group than in the LS group. Conclusion: The expression ofS100A9 and Galectin7 was associated with concurrent chemoradiotherapy sensitivity in cervical carcinoma.
