Abstract:
Objective: To study the effect of Uroacitide (CDA-2) on the proliferation, differentiation and autocrine ofhuman endometrial carcinoma cell line EAC.
Methods: MTT assay and colony formation assay were employed to detectthe effect of CDA-2 on cellular survival and proliferation. The differentiation of EAC cell line was identified by HE stain-ing and immunocytochemistry. E2 and P in endometrial carcinoma cell line EAC were quantified by radioimmunoassay.
Results: The cellular survival and proliferation of EAC cell line were remarkably inhibited by CDA-2 and the effect wasdose-dependent. After being treated with CDA-2 for 72 hours, HE staining of EAC cells revealed differentiation inducedby CDA-2. No expression of ER and PR were observed by immunocytochemistry. No E2 and P were detected in EACcells.
Conclusion: CDA-2 could inhibit proliferation and induce differentiation in human endometrial carcinoma cell lineEAC.
