Abstract:
Objective :To exam the proliferation and differentiation effect on human glioma cell line SWO-38 by Uroacitide (CDA-Ⅱ), which is a healthy human urine extraction.
Methods : The effect of CDA-Ⅱon cellular survival and colony formation was determined by MTT assay and colony formation assay. Then the anti-tumor effect of CDA-Ⅱwas examined by transplanting SWO-38 in nude mice in vivo. Finally, the change of cell morphology was observed by HE staining and light microscopy.
Results : After SWO-38 cells were treated with 1~5 mg/mL CDA-Ⅱ for 3 days, the inhibition rate of cell growth was 39.49%±5.27% to 65.05%±5.89%, the IC50 of CDA-Ⅱ was 2.52 mg/mL, and 10 days after the treatment with CDA-Ⅱ 0.3~2.1mg/mL, the inhibition rate of colony formation was 23.45%±0.62%~96.22%±1.01% and the IC50 was 1.03 mg/mL. Furthermore, the inhibitory effect was in a dose-dependent manner. CDA-Ⅱat dosage of 500 mg/kg/d and 2000 mg/kg/d with i.p. administration for 4 weeks could significantly suppress the growth of human glioma cells SWO-38 in nude mice, the inhibition rates were 47.77% and 79.94%, respectively (P<0.05, n=10=. HE staining and light microscopy revealed that CDA-Ⅱ could induce differentiation of human glioma cells SWO-38.
Conclusion : CDA- Ⅱdemonstrates a significant anti-tumor effect on human glioma and is a potential and natural anti-glioma drug.
