Abstract:
Objective :To detect the expression of c-Fos, and Cyclin D1 in Nasopharyngeal Carcinoma (NPC) and investigate the associatin between the expression, clinicopathological characteristics and the prognosis of NPC patients.
Methods : Archival formalin fixed, paraffin embedded specimens from 55 primary NPC patients, who had biopsy in Cancer center, Sun Yat-sen University during April in 1996 to June in 1998, were investigated. Immunohistochemistry LSAB method was adopted to detect the expression of c-Fos, Cyclin D1. The statistical analysis was performed using SPSS statistical software package (10.0).
Results : Positive staining for c-Fos was observed in cancer cell nucleus in 52 of 55 cases (94.5%). The expression of Cyclin D1 was positive in 47 of 55 cases (78.2%) and the staining site of Cyclin D1 was also in cancer cell nucleus. No significant association was observed among c-Fos, Cyclin D1 status and clinical stage, T stage, N stage, the sex and age. There was no statistically significant correlation between the c-Fos status and prognosis. Positive Cyclin D1 expression correlated with better OS and TTP. No significant correlation was found between Cyclin D1 status and DFS. On multivariate analysis, the age was more than 50 years and negative Cyclin D1 was an independent poor prognostic factors for NPC.
Conclusions :The Overexpression of c-Fos and Cyclin D1 has been observed inNPC and the negative Cyclin D1 is an independent poor prognostic factor for NPC.
