光敏剂photofrinⅡ预激活联合化疗药物对K562细胞杀伤作用的研究

The Studies on the Effect of Pre-activated Photofrin Ⅱ on K562 Cell Line Proliferation

  • 摘要: 目的: 通过体外研究探讨光敏剂photofrinⅡ预激活后对人白血病K562细胞株的杀伤作用及其与化疗药物配伍能否产生协同或拮抗效应. 方法: 以K562细胞株为研究对象,采用MTT法检测photofrinⅡ预激活后对K562细胞株的抑制作用,同时检测photofrinⅡ预激活后分别与顺铂(DDP)、5-氟脲嘧啶(5-FU)、阿霉素(ADM)联合应用对K562细胞的杀伤作用,Burgi法分析联合用药效果. 结果: photofrinⅡ预激活后能明显抑制K562细胞株的增殖,并且这种作用呈剂量依赖性,但其抑瘤作用较后激活组降低.photofrinⅡ预激活后分别与DDP、5-FU、ADM联合,增加了化疗药物的抑瘤作用. 结论: photofrinⅡ预激活后能够抑制K562细胞株的增殖,与DDP、5-FU、ADM联合使用具有相加作用.

     

    Abstract: Objective :To evaluate the influence of pactivated photofrin and it's combination with common anti-umor drubs on the growth of K562 cell line. Methods :K562 cell line was turgid in vitro with pactivated photofrin and different concentrations of DDP, 5-FU, or ADM.cule inhibition rate of K562 cell growth was evaluated by the method of MTT. Results :The pre-activated photofrin resulted in a dose lependent growth inhibition of K562 cell line, and enhanced the effect of c ytotoxicugs when used in combination. Conclusions :Preactivated photofrin inhibits the proliferation of K562 cell line. The combination of pre-activated photofrin and cytotoxicugs has a favorable effect.
