Abstract:
Objective : To investigate the effect of transforming growth factor(TGF )-β1 on proliferation in human Ovarian cancer cell line HO- 8910. Methods : Ovarian cancer cell line HO- 8910 was treated with TGF -β1 and assayed for growth response by MTT and FCM assay. Meanwhile, the expression level of endogenous Smad7, p15, p21 and of c-myc genes mRNA as well as the protein induced by TGF-β1 was detected using semi - quantitative RT - PCR and Western blot, respectively. Results : The proliferation of HO- 8910 cells was significantly inhibited by TGF- β1.After exposure to TGF-β1, the expression level of p15 and of Smad7 in cells was profoundly up- regulated and the level of c- myc was down - regulated. Conclusions : TGF-β1 may affect the growth inhibition through regulation of the expression levels of p15 and c- myc genes.
