转化生长因子-β1对人卵巢癌细胞增殖的抑制作用

Inhibitory Action of Transforming Growth Factor -β1 on Proliferation in Human Ovarian Cancer Cells

  • 摘要: 目的:探讨转化生长因子(TGF)-β1对人卵巢癌细胞系HO-8910增殖的影响及其作用机制。方法:采用MTT及流式细胞术检测TGF-β1对HO-8910细胞增殖的抑制作用,同时采用半定量RT-PCR和Westernblot方法分别检测TGF-β1作用不同时相细胞中Smad7、p15、p21和c-myc基因mRNA及蛋白表达水平的变化。结果:TGF-β1明显抑制HO-8910细胞的增殖;在TGF-β1(10ng/ml)刺激下,细胞中p15和Smad7的表达上调而c-myc的表达明显降低,p21的表达无明显变化。结论:TGF-β1可能通过上调p15、下调c-myc基因表达水平影响细胞周期调控,从而抑制卵巢癌细胞的增殖。

     

    Abstract: Objective : To investigate the effect of transforming growth factor(TGF )-β1 on proliferation in human Ovarian cancer cell line HO- 8910. Methods : Ovarian cancer cell line HO- 8910 was treated with TGF -β1 and assayed for growth response by MTT and FCM assay. Meanwhile, the expression level of endogenous Smad7, p15, p21 and of c-myc genes mRNA as well as the protein induced by TGF-β1 was detected using semi - quantitative RT - PCR and Western blot, respectively. Results : The proliferation of HO- 8910 cells was significantly inhibited by TGF- β1.After exposure to TGF-β1, the expression level of p15 and of Smad7 in cells was profoundly up- regulated and the level of c- myc was down - regulated. Conclusions : TGF-β1 may affect the growth inhibition through regulation of the expression levels of p15 and c- myc genes.
