Abstract:
Objective: To investigate the molecular mechanism of the effect of A3D8, an antiCD44 monoclonal antibody on proliferation and differentiation of human HL-60 cells. Methods: By FCM, RT-PCR and Western blot, the expression of Cyclin D1, CDK4 and p21cip1 was determined in HL-60 cells, after ligation with A3D8 anti-CD44 mAb. Results: A3D8 anti-CD44 mAb induces a G

1 arrest by upregulating the expression of p21cip1 and downregulating the expression of Cyclin D1 and CDK4 in HL-60 cells. Conclusions: Our results demonstrate that A3D8 anti-CD44 mAb inhibits proliferation and induces differentiation of HL-60 cells. Its molecular mechanism may involve the increase of p21cipl expression and inhibition of cell cycle regulatory proteins Cyclin D1 and CDK4 expression.
