The Effect of A3D8 Anti-CD44 Monoclonal Antibody on Expression of Cyclin D1, CDK4 and p21cip1 of Human HL-60 Cells

  • 摘要: 目的:探讨CD44单克隆抗体A3D8对人急性髓系白血病细胞株HL-60细胞增殖分化影响的分子作用机制。方法:采用FCM、RT-PCR及Westernblot方法检测A3D8作用前后HL-60细胞CyclinD1、CDK4、p21cip1表达的变化。结果:A3D8作用使HL-60细胞发生G0/G1期阻滞。A3D8下调HL-60细胞CyclinD1及CDK4的表达,上调HL-60细胞p21cip1mRNA及P21蛋白表达。结论:A3D8抑制HL-60细胞增殖、诱导其分化的分子机制可能与p21cip1表达上调及CyclinD1、CDK4表达下调有关。

     

    Abstract: Objective: To investigate the molecular mechanism of the effect of A3D8, an antiCD44 monoclonal antibody on proliferation and differentiation of human HL-60 cells. Methods: By FCM, RT-PCR and Western blot, the expression of Cyclin D1, CDK4 and p21cip1 was determined in HL-60 cells, after ligation with A3D8 anti-CD44 mAb. Results: A3D8 anti-CD44 mAb induces a G0/ G1 arrest by upregulating the expression of p21cip1 and downregulating the expression of Cyclin D1 and CDK4 in HL-60 cells. Conclusions: Our results demonstrate that A3D8 anti-CD44 mAb inhibits proliferation and induces differentiation of HL-60 cells. Its molecular mechanism may involve the increase of p21cipl expression and inhibition of cell cycle regulatory proteins Cyclin D1 and CDK4 expression.
