马鞭草C部位诱导人绒毛膜癌JAR细胞凋亡分子机制研究

The Studies on the Part C of Verbena Officinalis-induced Human Choriocarcinoma JAR Cells Apoptosis and Its Molecular Mechanism in vitro

  • 摘要: 目的: 探讨马鞭草C部位诱导人绒毛膜癌JAR细胞凋亡的分子机制。 方法: 流式细胞仪检测细胞周期;Western-blot及RT-PCR检测Fas/Fasl蛋白及基因的表达。 结果: 药物作用后细胞被不同程度阻滞于G2/M期,并伴有G0/G1期比例下降;Fasl蛋白表达降低,Fas在C部位作用前后均无表达;FaslmRNA转录呈下降趋势。 结论: 马鞭草C部位将人绒毛膜癌JAR细胞阻滞在G2/M期,并诱导其凋亡,其作用机制可能与Fasl的表达下调有关。

     

    Abstract: Objective :To investigate the biological effect of part C and the potential mechanisms on Human Choriocarcinoma JAR Cell Line. Methods :FCM was performed for measurement of phase of cell cycle; the expressions of Fas and Fasl wire determined by Western-blot and RT- PCR. Results :Verbena officinalis could induce a G2 /M phase arrest to the different degrees in this cell line with the GO /G1 phase proportions down- regulated. Western- blot and RT- PCR analysis showed that Fasl proteinand Fas1mRNA were down-regulated after treatment, while Fas was not expressed before and after it. Conclusions :G2/M phase arrest is one of the mechanisms of part C-induced Apoptosis. Fasl is involved in the process and its mechanisms may be reducing the expression of Fas1mRNA.
