The Over-expression of Pin1 and Its Association with Ki67 in Cervical Cancer

  • 摘要: 目的:探讨宫颈癌中肽基脯氨酰顺反异构酶Pin1表达、与Ki67的相关性及其临床意义。方法:应用RT-PCR和免疫组化技术检测宫颈癌组织中Pin1mRNA和蛋白表达水平及其与Ki67蛋白表达的相关性。结果:1)宫颈癌组织中Pin1mRNA表达明显高于正常宫颈组织(P<0.05);2)从正常宫颈到CIN再到浸润癌Pin1蛋白表达逐渐增强(P<0.05);3)Pin1蛋白过表达与临床分期、病理分级、淋巴结转移无明显相关(P>0.05);但宫颈腺癌Pin1表达明显高于宫颈鳞癌(P<0.05);4)Pin1过表达与Ki67表达呈显著正相关(P<0.05)。结论:1)Pin1过表达与宫颈癌细胞增殖有关,参与宫颈癌发生发展;2)Pin1过表达可作为宫颈癌诊断的辅助指标。

     

    Abstract: Objective :To study the expression status of Pin1,its association with Ki67 and their clinical importance in cervical cancer. Methods :Amplification of Pin1 gene was examined by RT-PCR,and the expression of both Pin1 and Ki67 protein were examined by immunohistochemistry in cervical cancer tissues. Results :1.A significantly higher expression of Pin1 mRNA was detected in cervical cancer than those in normal cervical tissues(P<0.05).2.The expression of Pin1 increased progressively along with the disease process from normal cervix to CIN and to cervical cancer(P<0.05).3.No significant difference of Pin1 expression was found between disease stage(FIGO),pathological grade or pelvic lymph node metastasis status(P>0.05).But the expression of Pin1 was significantly higher in adenocarcinoma than those in squamous carcinoma of the uterine cervix(P<0.05).4.In cervical cancer,the overexpression of Pin1 was positively correlated with that of Ki67(P<0.05). Conclusion :The overexpression of Pin1 is closely related to cancer cell proliferation or progression of cervical cancer and contributed to oncogenesis.Pin1 may serve as a potential marker for cervical cancer diagnosis.
