补骨脂素对HL60细胞凋亡及细胞内Ca2+浓度影响的探讨

Apoptosis of HL60 Cell Line Induced by Psoralean and Its Effect on Intracellular Ca2+ Concentration

  • 摘要: 目的:探讨补骨脂素对HL60细胞凋亡的影响,以及细胞内Ca2+浓度与凋亡之间关系。方法:采用MTT比色分析法测定补骨脂素对HL60的细胞毒性影响,流式细胞仪及激光共聚焦显微镜测定HL细胞内Ca2+浓度和凋亡比率。结果:补骨脂素对HL60细胞生长具有抑制作用,补骨脂素促进细胞凋亡比率,细胞凋亡与Ca2+浓度成正相关。结论:补骨脂素影响HL60细胞凋亡,Ca2+介导的细胞凋亡是补骨脂素抑制肿瘤细胞生长的可能机制之一。

     

    Abstract: Objective: To investigate the apoptosis induced by Psoralean, and the relationship between intracellular Ca2+ Concentration and apoptosis. Methods: MTT assay, Annexin V FITC and laser scanning confocal microscopy were used to study the inhibitory effect of HL60 cells on cell proliferation and the induction of apoptosis ratio in HL60 cells. Results: Psoralean can inhibit the growth of HL60 cells, increasing the ratio of apoptosis, Ca2+Concentration and apoptosis is positive correlation. Conclusion: Psoralean can influence HL60 cells apoptosis and the Ca2+-induced apoptosis is one of the possible mechanisms of Psoralean inhibiting the growth of cancer cell.
