粉防已碱对人卵巢癌细胞株A2780增殖与凋亡的影响

The Effects of Tetrandrine on Proliferation and Apoptosis of Hunman Ovarian Cancer Cell Line A2780

  • 摘要: 目的:通过观察粉防已碱对人卵巢癌A2780细胞增殖与凋亡的影响,初步探讨粉防已碱对卵巢癌的体外抗肿瘤效应。方法:采用MTT试验观察粉防已碱对A2780细胞的增殖抑制效应,利用细胞DNA琼脂糖凝胶电泳检测粉防已碱诱导肿瘤细胞凋亡的作用,通过流式细胞术观察粉防已碱对人卵巢癌A2780细胞周期的影响以及诱导凋亡的作用,以免疫印迹检测粉防已碱对p53、p21及Bax表达水平的影响。结果:MTT试验显示粉防已碱对人卵巢癌A2780细胞增殖有抑制作用,其抑制效应具有时间及浓度依赖的特点,琼脂糖凝胶电泳及流式细胞术表明粉防已碱可诱导A2780细胞凋亡,并引起G0/G1期阻滞,免疫印迹检测显示粉防已碱上调p53、p21及Bax蛋白表达。结论:粉防已碱对人卵巢癌A2780细胞增殖的抑制效应具有时间及浓度依赖性,并可诱导细胞凋亡,其抗肿瘤效应可能与上调p53、p21及Bax蛋白表达有关。

     

    Abstract: Objective :To investigate the anti-tumor effect of tetrandrine on human ovarian cancer in vtro,effects of tetrandrine on proliferation and apoptosis of human ovarian cancer cell A2780 were observed. Methods :The effect of tetrandrine on proliferation of A2780 cells was observed by MTT assay.Detection of apoptosis and cell cycle analysis were performed with flow cytometry,and DNA fragmentation was verified by agarose electrophoresis.The expressions of p53,p21 and Bax were analyzed by Western blot. Results :Tetrandrine inhibits the proliferation of A2780 cells in dosage and time dependent manner and induces apoptosis.The cell cycle was arrested in G0/G1 phase in A2780 cells treated with tetrandrine.Western blot showed that the expressions of p53,p21 and Bax increase in A2780 cells treated with tetrandrine. Conclusion :Tetrandrine inhibits the proliferation of A2780 in the dose and time dependent manner,and induces apoptosis.The anti-tumor effect of tetrandrine may be due to up-regulating the expressions of p53,p21 and Bax.
