尼美舒利对人食管癌Eca-109细胞凋亡及Survivin和Caspase-3表达的影响

The Effect of Nimesulide on Apoptosis and Expression of Survivin and Caspase-3 on Human Esophageal Carcinoma Eca-109 Cells

  • 摘要: 目的:观察环氧化酶-2(COX-2)选择性抑制剂尼美舒利对人食管癌Eca-109细胞增殖及凋亡的影响,研究其对凋亡抑制蛋白Survivin和Caspase-3表达的影响,探讨尼美舒利诱导Eca-109细胞凋亡的作用机制。方法:尼美舒利作用Eca-109细胞后,MTT法测定尼美舒利对人食管癌Eca-109细胞增殖的抑制率;电子显微镜和流式细胞仪检测细胞凋亡;RT-PCR法检测Eca-109细胞SurvivinmRNA表达变化,Westernblot检测Survivin和Caspase-3蛋白表达变化。结果:尼美舒利(50~400μmol/L)对Eca-109细胞生长有抑制作用,随浓度升高、时间延长抑制作用增强,并诱导Eca-109细胞凋亡,呈剂量-时间效应关系;尼美舒利可降低SurvivinmRNA和蛋白表达,增加Caspase-3蛋白表达。结论:尼美舒利可诱导人食管癌细胞株Eca-109凋亡,其机制可能与下调Survivin表达及激活Caspase-3表达有关。

     

    Abstract: Objective: To detect the effect of nimesulide, the selective inhibitor of cyclooxygenase-2 (COX-2), on the cell proliferation and apoptosis of human esophageal carcinoma Eca-109 cell lines, study the expression of Survivin and Caspase-3, and to investigate the mechanism of nimesulide to induce tumor cell's apoptosis. Methods: After the treatment with nimesulide on esophageal carcinoma Eca-109 cell, MTT assay was used to observe the inhibition of Eca-109 cell growth and apoptosis was determined by electronic microscopy and flow cytometry. The expression of Survivin mRNA was detected by RT-PCR and the protein expression of Survivin and Caspase-3 was examined using Western blot. Results: The dose of 50~400μmol/L of nimesulide showed the inhibitory effect on growth of Eca-109 cell lines and induce apoptosis in a dose- and time-effective manner, moreover nimesulide also down-regulated the level of mRNA and protein of survivin, and elevated the expression of capase3. Conclusion: Nimesulide could induce apoptosis of the esophageal carcinoma Eca-109 cells by down-regulation of expression of survivin and up-regulation of caspase-3.
