Abstract:
Objective :The study was designed to investigate the effects of CD151 on proliferationof Human Cervix Carcinoma Cell Line Hela.
Methods : The recombinant vectors pcDNA3.1-CD151,pcDNA3.1-antiCD151 and pcDNA3.1-GFP were respectively transfected into the Hela cell line byLipofectamine TM 2000 transfection. Protein level of CD151 in Hela cells was detected by Western blot.MTT assay, flow cytometry and the analysis of PCNA expression were performed to measure the effectsof CD151 on Hela cells proliferation.
Results : Transfection of pcDNA3.1-CD151 increased expressionof CD151 and PCNA(P<0.01),the cell proliferation rate in MTT assay (P<0.01) and the cell proportionin S+G
2/M phage (P<0.05) in Hela cells, and accelerated cell growth and proliferation. On the con-trary,transfection of antisense CD151 gene decreased expression of CD151 and PCNA(P<0.01), the cellproliferation rate in MTT assay (P<0.01) and the cell proportion in S+G
2/M phage (P<0.05) in Helacells,and suppressed cell proliferation.
Conclusion :CD151 has an effective role in proliferation in tu-mor cells. Through the intervention of antisense CD151 gene transfection, the expression of CD151 canbe down-regulated significantly and thereby suppressed tumor cell proliferation.
