纤维粘连蛋白mRNA表达量与乳腺癌转移的关系及临床意义

The Clinical Significance and Relationship between Expression of Fibronectin mRNA and Metastasis of Breast Cancer

  • 摘要: 目的:探讨乳腺癌纤维粘连蛋白(Fibronectin,FN)mRNA表达量与乳腺癌转移的关系及临床意义。方法:采用荧光定量RT-PCR方法,检测30例乳腺原发癌和配对淋巴结转移癌以及116例乳腺原发癌的FNmRNA表达量,比较30例乳腺原发癌和淋巴结转移癌的FNmRNA表达量,分析FNmRNA表达量与乳腺癌临床病理因素的关系。结果:30例乳腺淋巴结转移癌FNmRNA表达量低于原发癌(P<0.05)。肿瘤﹥5cm的FNmRNA表达量低于肿瘤≤5cm(P<0.05),临床期的FNmRNA表达量低于Ⅰ期和Ⅱ期(P<0.05),淋巴结4枚以上阳性FNmRNA表达量低于1~3枚阳性(P<0.05),雌、孕激素受体阳性的FNmRNA表达量高于雌、孕激素受体阴性。FNmRNA表达量与病理类型、组织学分级、Her-2表达、CA153表达无关。结论:FNmRNA表达量在临床早期及淋巴结转移早期上调,随着肿瘤的进展表达量下调。FNmRNA转录水平的表达与乳腺癌转移有关。

     

    Abstract: Objective: To discuss the clinical significance and relationship between expression of fibronectin mRNA and metastasis of breast cancer. Methods: The fibronectin mRNA expression of 30cases with paring lymph-node metastatic carcinoma and 116 with primary breast cancer were detectedand the relationship between the fibronectin mRNA expression and clinicopathologic factors of breast cancer was analyzed using fluorescent-quantitative RT-PCR. Results: FN mRNA expression in 30metastatic lymphnode was lower than in primary breast cancer (P<0.05). The mRNA expression of FNin the tumor over 5cm was lower than in those less or equal to 5cm (P<0.05). The FN mRNA expres-sions in stageⅠ and were lower than in stage Ⅱ (P<0.05). The mRNA expression of FN in more than 4lymph nodes positive was lower than in lymph node 1-3 positive (P<0.05). The mRNA expressions of FN in ER and PR positive were higher than in ER and PR negative. The FN mRNA expression was ir-related with tissue grade, pathologic type, Her-2 and CA153. Conclusion: FN mRNA expression isup-regulated in the clinical early stage of the lesion and the early stage of lymph-node metastasis, andis down-regulated with the progression of tumors. FN mRNA expression is related with the metastasis of breast cancer.
