塞来昔布诱导Ls174结肠腺癌细胞周期阻滞和凋亡

Induction of Cell Cycle Arrest and Apoptosis by Celecoxib in Ls174 Cells

  • 摘要: 目的 :观察选择性COX-2抑制剂塞来昔布(Celecoxib)诱导结肠腺癌细胞株Ls174凋亡的作用及机制。 方法 :采用MTT法检测塞来昔布对Ls174细胞的增殖抑制作用,应用流式细胞仪检测塞来昔布对Ls174细胞凋亡和增殖周期的影响,并检测塞来昔布对Ls174细胞Caspase-3活性的影响。 结果 :塞来昔布抑制Ls174细胞增殖和诱导Ls174细胞的凋亡作用均呈剂量和时间依赖性,塞来昔布处理后的Ls174细胞G0/G1期百分比与对照组相比明显增高。塞来昔布能诱导Ls174细胞Caspase-3活性,并呈浓度和时间依赖性。 结论 :塞来昔布在体外能抑制Ls174细胞的增殖,诱导细胞凋亡,其作用与促进Caspase-3活性和阻滞细胞周期于G0/G1期有关。

     

    Abstract: Objective :To study the anti-tumor mechanism of Celecoxib and to investigate its ef-fect on cell cycle and apoptosis in Ls174 cells. Methods :Cell growth inhibition of Celecoxib was mea-sured by MTT assay;the induction of cell cycle arrest and apoptosis by Celecoxib was determined byflow cytometry. Caspase-3 activity was detected using a colorimetry assay. Results :Celecoxib displayedstrong growth inhibition on Ls174 cells in a dose-and time-dependent manner.Ls174 cells were in-duced to undergo apoptosis and underwent G0/G1 arrest with exposure to Celecoxib. Celecoxib marked-ly increased caspase- 3 activities of Ls174 cells. Conclusion :Celecoxib-induced cell cycle arrest andapoptosis may play an important role in its anti-tumor effect,and Celecoxib-induced apoptosis is close-ly related to enhancement of Caspase-3 activity.
