Role of CD44v3 in the In Vitro Adhesion and Invasion of Hyaluronic acid-Induced Human Lung Carcinoma Cell A549

  • 摘要: 目的:研究CD44v3在透明质酸诱导A549细胞体外粘附和侵袭中的作用。方法:设计和构建能产生针对CD44v3的短发夹RNA(shorthairpinRNA,shRNA)的RNA干扰质粒,转染A549细胞,以RT-PCR和Westernblot检测A549细胞转染前后CD44v3表达,以平板粘附模型和波登氏小室模型检测A549细胞转染前后粘附和侵袭能力。结果:和转染前相比,转染了RNA干扰质粒的A549细胞CD44v3mRNA和蛋白表达以及受透明质酸诱导而粘附于平板和穿过波登氏小室隔膜的细胞数皆明显减少(P<0.01)。结论:CD44v3介导了透明质酸诱导人肺腺癌A549细胞体外粘附和侵袭行为。

     

    Abstract: Objective: To investigate the role of CD44v3 in adhesion and invasion behavior of human lung carcinoma cell A549 induced by hyaluronan in vitro. Methods: The RNA interference plasmid expressing shRNA (short hairpin RNA) against CD44v3 was designed and constructed, and was used for transfection of the A549 cell line. The expression of CD44v3 before and after transfection of the A549 cell line was examined by the reverse transcription polymerase chain reaction (RT-PCR) and western blot methods. Both Adhesive and invasive ability before and after transfection of the A549 cell line were determined by the plate adhesion model and Boyden chamber model. Results: Compared to pre-transfection, the A549 cell CD44v3 mRNA and expression of proteins transfected with RNA interference plasmid as well as the cell population sticking to the flat plate and penetrating septum of Boyden chamber decreased significantly (P<0.01). Conclusions: CD44v3 mediates the in vitro adhesion and invasive behavior of hyaluronic acid-induced human lung carcinoma cell line A549.
