PPO, a New Putative Oncogene in MAPK Signaling Pathway

  • 摘要: 目的:在1p36区寻找新的癌基因,用于癌症的诊断和治疗。方法:通过克隆筛选与癌症相关的基因。定量PCR检测该基因在各种肿瘤组织和细胞株中的表达量。利用基因重组的方法构建该基因的表达质粒,并研究该基因在细胞水平和活体水平对细胞增殖及磷酸化功能的影响。结果:在人类第1号染色体上发现了一个新的癌基因PPO。人类多种肿瘤组织中,该基因均呈高表达,从细胞水平到活体水平均证实,该基因与增殖和磷酸化有关。结论:我们在1p36区克隆出了一个新的基因,该基因与细胞增殖及磷酸化功能有关。并命名为PPO(ProliferationandPhosphorylationOncogene)。PPO能使MAPK通路中ERK2和MEK1/2磷酸化,推测PPO可能是在MAPK通路中一个新的癌基因。

     

    Abstract: Objective: To find a new putative oncogene for diagnosis and treatment of the cancer in the 1p36 region. Methods: A new putative oncogene was identified by screening the 1p36 region. The quantity of expression of different cancer tissues and cell lines were detected using the real time RT-PCR. The expression plasmid was constructed by recombination of the gene. The function of proliferation and phosphorylation in vitro or in vivo was further studied. Results: After screening the 1p36, a new putative oncogene PPO was identified. It was over-expressed in different kind of cancer tissues and cell lines. The function of PPO was related with proliferation in vitro and in vivo both. The phosphorylation status of the ERK2 and MEK1/2 was up-regulated. Conclusion: The PPO is a newputative oncogene in MAPK signaling pathway.
