The Effect of KAI1 Gene on Biological Behavior of Breast Cancer Cells

  • 摘要: 目的:研究稳定转染外源性KAI1基因对人乳腺癌细胞体外生物学特性的影响。方法:应用脂质体将pCMV-KAI1质粒转染入人乳腺癌细胞株MDA-MB-231中,免疫印迹方法检测蛋白表达,分别利用MTT法,体外粘附及侵袭力实验判断KAI1基因对细胞增殖能力及粘附,侵袭力的影响,并利用ELISA法检测转染前后游离组织间粘附因子-1(sICAM-1)的动态变化。结果:获得了稳定表达KAI1基因的MDA-MB-231乳腺癌细胞克隆,并有KAI1蛋白的高表达。MTT法显示转染组的细胞增殖力低于未转染组和转染空载体组(P<0.05)。体外粘附及侵袭力实验表明转染组的细胞粘附侵袭能力低于未转染组和转染空载体组(P<0.05)。转染后sICAM-1含量下降。结论:KAI1基因可抑制人乳腺癌细胞的恶性特征,并可能是通过影响一些粘附因子作用而抑制肿瘤细胞转移。

     

    Abstract: Objective: To investigate the effect of stable transfection of the exogenous wild KAI1 gene on biological behavior of the breast cancer cell in vitro. Methods: Human breast cancer cell lines MDA-MB-231 was transfected with pCMV-KAI1 or mock transfected plasmid pCMV with lipofectamine. Western blot was used to determine the expression of target protein of KAI1. The proliferative ability of cells was tested by MTT assay and the metastatic ability was investigated by cell adhesion and invasion assay. The contents of ICAM-1 before and after the transfection were also measured using ELISA assay. Results: The MDA-MB-231 breast cancer cell clone with stable expression of KAI1 gene was acquired and a high expression of the KAI1 protein was observed. The MTT showed cell proliferation ability was lower in the transfection group than in the non-transfection and mock-vehicle groups (P<0.05). Experiments on the in vitro adherency and invasiveness showed that cell adhesion and invasion ability was lower in the transfection group compared to that in the non-transfection and mock-vehicle groups (P<0.05). The content of sICAM-1 decreased after the transfection. Conclusion: The KAI1 gene can restrain malignant phenotypes of the human breast cancer cells in vitro and may inhibit the metastasis by influencing the function of some adherence factors.
