细胞周期调控因子与下咽癌生物学行为相关性初步研究

Pr eliminary Study of the Relationship between Cell Cycle Regulatory Factor s and Biological Behavior of Hypopharyngeal Car cinoma

  • 摘要: 目的:探讨细胞周期调控因子与下咽癌临床生物学行为之间的关系。方法:取下咽鳞状细胞癌手术标本53例,癌旁正常粘膜标本11例,应用免疫组化SP法检测P27Kip1、CyclinD1、CDK4在下咽癌组织及正常粘膜中的表达。结果:P27Kip1、CyclinD1、CDK4均为弥漫性细胞核表达。P27Kip1在下咽癌中的表达低于正常粘膜,且表达与N分级明显相关(P<0.05);CyclinD1在下咽癌中的表达高于正常粘膜,且表达与T分级明显正相关(P<0.05);CDK4在下咽癌中的表达高于正常粘膜,且表达与T分级、N分级明显正相关(P<0.01、P<0.05)。P27Kip1与CyclinD1,P27Kip1与CDK4在下咽癌组织中多呈反向表达,CyclinD1与CDK4多呈同向表达。结论:CyclinD1、CDK4与P27Kip1在下咽癌发病机制中共同作用,对预后有一定的参考价值。

     

    Abstract: To determine the relationship between cell cycle regulatory factors and biological behavior of hypopharyngeal squamous cell carcinoma. Methods: Immunohistochemical staining by specific monoclonal anti- p27/Kip1 protein, anti- cyclin D1 protein and anti- CDK4 protein was performed using the S- P method in 53 cases of hypopharyngeal squamous cell carcinoma and 11 cases of normal epithelium. Results: All of the positive staining for Cyclin D1, CDK4 and p27/Kip1 in hypopharyngeal squamous cell carcinoma showed suffusing expression in the nucleolus. There was a significant difference in expression between the cancerous cells and normal epithelium. The negative lymph node group had a significantly higher immunoreactivity for p27/Kip1 than the lymph node positive group (P<0.05). Cyclin D1 expression was related to T grade (P<0.05). A significant positive correlation was found between CDK4 expression and T and N grade (P<0.01, P<0.05). The reverse pattern was found in p27/Kip1- Cyclin D1 and p27/Kip1- CDK4. On the other hand, unidirectional expression was found for Cyclin D1 and CDK4. Conclusion: Cyclin D1, CDK4 and p27/Kip1 each play a role in hypopharyngeal squamous cell carcinoma progression and together can predict the probable disease course and outcome.
