TRF1和TANK1在非小细胞肺癌中的异常表达及其临床意义

Abnormal Expr ession and Clinical Significance of Telomer ic Repeat Binding Factor 1 and Tankyr ase 1 in Non- small Cell Lung Cancer

  • 摘要: 目的:研究端粒结合蛋白TRF1(telomeric repeat binding factor1)和端锚蛋白TANK1(tankyarse1)在非小细胞肺癌(non-small cell lung cancer,NSCLC)中的表达,探讨TRF1mRNA与NSCLC的组织类型、病理分级、TNM分期及淋巴结转移的关系。方法:采用RT-PCR对40例NSCLC患者癌组织、癌旁组织的TRF1、TANK1mRNA进行半定量检测。40例患者中20例为腺癌,20例为鳞癌。结果:癌组织TRF1mRNA水平比癌旁组织明显下降,并且低分化癌组织中的TRF1mRNA水平比高分化癌组织中明显下降,差异均有显著性(P<0.05);癌组织TANK1mRNA表达水平比癌旁组织明显增加,差异有显著性(P<0.05)。结论:NSCLC组织中TRF1mRNA呈低表达;而TANK1mRNA呈高表达;TRF1mRNA的表达水平与组织分化程度相关。

     

    Abstract: To investigate the expression level of mRNA encoded by the TRF1 (telomeric repeat binding factor 1) and tankyrase 1 (TANK1 ) in cancer tissues and paired noncancerous tissue. Methods: A total of 40 patients including 29 males and 11 females with non- small cell lung cancer (NSCLC) underwent radical surgery. There were 20 cases of squamous cell carcinoma and 20 cases of adenocarcinoma in the group. The expression of TRF1 and TANK1 mRNA was measured by RTPCR. Results: TRF1 mRNA was significantly downregulated in cancer tissue compared to the paired noncancerous tissue (P<0.05). TANK1 mRNA was significantly upregulated in cancer tissue compared to the paired noncancerous tissue (P<0.05). In addition, the expression of TRF1 mRNA was significantly associated with the grade of tumor differentiation (P<0.05). No significant difference in the TRF1 mRNA level was found between the patients of different sexes or among different clinical stages, pathological subtypes and lymph node metastasis (P>0.05). Conclusions: Downregulation of TRF1 mRNA expression and upregulation of TANK1 mRNA expression occur in lung cancer tissue. TRF1 may play a significant role in cell differentiation in NSCLC.
