分化相关基因MIBP1的表达与人脑胶质瘤恶性程度的关系

The Relationship between Expr ession of MIBP1, a Tumor Differ entiation- associated Gene, and Malignancy of Human Glioma

  • 摘要: 目的:应用荧光定量实时PCR技术检测与肿瘤分化相关的基因MIBP1(c-myc内含子结合蛋白1)在不同恶性程度的人脑胶质瘤中的表达。方法:收集病理确诊的30例不同恶性程度胶质瘤以及5例正常脑组织手术标本,抽提RNA,应用荧光定量PCR技术检测MIBP1基因在人脑胶质瘤中的表达,并分析该基因在不同级别胶质瘤中的表达情况。结果:荧光定量PCR系统稳定,WHOⅣ级胶质瘤与正常对照及Ⅱ级胶质瘤比较有显著性差异,多形性胶质母细胞瘤MIBP1基因表达明显降低。2例自身对照标本分析,Ⅳ级肿瘤中表达亦降低。结论:c-myc调控基因MIBP1在高度恶性胶质瘤中表达受抑制,提示该基因的下调可能为癌变的晚期事件。MIBP1基因可能有抑癌作用。

     

    Abstract: To study the expression of MIBP1, the differentiation- associated gene also known as c-myc intron binding protein 1, in specimens of cerebral glioma using real-time PCR. Methods: Tissues from 30 cases of glioma and 5 samples of normal human brain were collected after neurosurgical operations and were grouped based on pathological results. Total RNA was extracted and expression of MIBP1 mRNA was quantified by real- time PCR according to the manufacturer' s recommended protocol. The differences in expression level among the various groups were studied. Results: The real-time PCR system showed a high reliability for our particular reactions. The expression of MIBP1 in the WHO grade- IV glioma was obviously lower compared to the normal controls and all other groups. Conclusion: MIBP1, a c- myc regulating gene, shows decreased expression in gliomas of high malignancy, suggesting that downregulation of the MIBP1 gene might be a late event in oncogenesis of glioma. MIBP1 is a probable candidate for a tumor suppressor gene.
