肿瘤骨转移对外周血造血干细胞采集影响的初步观察

Preliminary Observation on Influence of the Osseous Metastasis of Tumor on Autologous Peripheral Blood Stem Cell Collection

  • 摘要: 目的:探讨肿瘤骨转移对接受外周血造血干细胞采集的影响。方法:选择2004年4月至2006年6月接受自体外周血造血干细胞移植的患者共36例,根据有无肿瘤骨转移分为两组,A组合并肿瘤骨转移;B组无肿瘤骨转移。两组均采用泰素帝120mg/m2联合G-CSF5μg/kg/日动员外周造血干细胞,血细胞分离机采集单个核细胞。检测采集前外周血CD34+细胞比例和采集物单个核细胞、CD34+细胞数量,评价两组在动员效果和采集物质量方面的差异。结果:A、B两组动员后外周血单个核细胞分别为39.3±14.7%、41.1±12.4%;CD34+细胞比例分别为0.16±0.07%、0.17±0.10%。A、B两组采集物的单个核细胞数量分别为3.47±1.16×108/kg、4.02±1.31×108/kg;CD34+细胞数量分别为2.52±1.43×106/kg、2.73±1.87×106/kg。经检验A、B两组动员后外周血和采集物的CD34+细胞数量间无统计学差异。结论:肿瘤骨转移作为单因素可能对自体外周血造血干细胞的动员和采集质量无影响。

     

    Abstract: Objective: To discuss the influence of tumor osseous metastasis on the patients receiving autologous peripheral blood stem cell collection. Methods: A total of 36 patients with malignant diseases received the autologous peripheral blood stem cell transplantation, during a period from April 2004 to June 2006, were chosen. The patients were divided into two groups, i.e. the group A for patients with a complication of tumor osseous metastasis and group B without the metastasis. Both groups were administrated with Taxotere 120mg/m2 plus granulocyte colony- stimulating factor(G- CSF) 5μ g/kg/d, the mobilization regimen. Blood cell separator was used to collect the mononuclearcells. The proportion of harvested CD34+ cells in peripheral blood, the collected monoclearcells were detected by Flow Cytometer and the numbers of CD34+ cells calculated accordingly to appraise the difference between effect of the mobilization and quality of the collections of the two groups. Results: After the mobilization in the group A and B, the number of the peripheral blood mononuclear cells (PBMC) was 39.3±14.7% and 41.1± 12.4% and the proportion of CD34+ cells was 0.16± 0.07% and 0.17± 0.10%, respectively. After the tests, there was no significant difference between the number of harvested PBMC and CD34+ cells of the two groups, i.e., 3.47± 1.16× 108/kg and 2.52± 1.43× 106/Kg in group A and 4.02±1.31× 108/kg and 2.73± 1.87× 106/Kg in group B, respectively. Conclusion: Osseous metastasis, as a single factor, may have no impact on mobilization and harvesting of the hematopoietic stem cells and their engraftment after autotransplantation.
