乙肝病毒基因产物和HSP70在肝炎相关性肝癌发生中的作用

Effects of HBV Gene Products and HSP70 on Development of Hepatitis- related Hepatocellular Carcinoma

  • 摘要: 目的:探讨乙肝病毒基因产物(HBsAg,HBcAg,HBxAg)和热休克蛋白70(hot shock protein70,HSP70)在乙型肝炎相关性肝癌中的表达及其在肝细胞癌变中的作用。方法:采用组织芯片技术和免疫组织化学染色方法对152例乙型肝炎相关肝细胞肝癌组织、78例乙型肝炎后肝硬化组织中HSP70、HBsAg、HBcAg和HBxAg的表达情况进行检测。结果:与乙型肝炎后肝硬化相比,在乙型肝炎相关性肝癌组织中HBsAg表达显著降低(P=0.000),HBcAg表达显著增高(P=0.000),HBxAg显著增高(P=0.005),HSP70呈现高表达(P=0.000);相关分析结果显示,HSP70的表达和HBcAg的表达呈显著正相关(P=0.009),而和HBxAg的表达未显示相关性(P=0.673)。结论:乙肝病毒抗原成分HBcAg、HBxAg以及HSP70参与在乙型肝炎相关性肝癌的发生过程,HBcAg可能通过增加HSP70的表达而参与肝癌的发生。

     

    Abstract: Objective: To study the expression of the products of the HBV genome and HSP70 in hepatitis- related hepatocellular carcinomas and to determine the possible mechanism in the course of carcinogenesis. Methods: Samples from a total of 152 cases with carcinoma and 78 cases with post-hepatitis liver cirrhosis were stained immunohistochemically in tissue array format to detect expression of the HBV antigens HBsAg, HBcAg, HBxAg and HSP70. Results: The expression levels of HBcAg,HBxAg, and HSP70 were significantly higher in carcinoma than in cirrhosis (P=0.000, P=0.005 and P=0.000, respectively). The expression of HBsAg was significantly lower in carcinoma than in cirrhosis (P=0.000). Correlation analysis showed that the expression of HSP70 was significantly correlated with the expression of HBcAg (P=0.009) but was not related to the HBxAg (P=0.673). Conclusion: HBcAg,HBxAg and HSP70 participate in the development of hepatitis - related hepatocellular carcinoma;HBcAg may take part in such a course by a mechanism related to HSP70. HBcAg may be involved in carcinogenesis through an increase in HSP70 expression.
