siRNA干扰对Hela细胞株VEGF表达的影响

Effect of Small interference RNA on Expression of the Vascular Endothelial Growth Factor in Hela Cell Line

  • 摘要: 目的:探讨血管内皮生长因子(VEGF)siRNA干扰对宫颈癌Hela细胞株VEGF的影响。方法:设计合成VEGFsiRNA1、2、3号链,体外脂质体介导转染宫颈癌Hela细胞株,半定量RT-PCR及Western blot分析转染效果。结果:转染VEGF siRNA1、2号链后,宫颈癌Hela细胞株VEGFmRNA表达明显下降,但是VEGF蛋白水平下降不明显。结论:VEGF siRNA转染宫颈癌Hela细胞株可下调细胞中VEGFmRNA的表达,不同的siRNA下调mRNA效率不同。作为基因沉默的一个重要工具,siRNA干扰有望成为治疗宫颈癌一种新方法。

     

    Abstract: Objective: To study the inhibition of Vascular Endothelial Growth Factor (VEGF) by Small Interference RNA (SiRNA) on the effect of VEGF in Hela cell line of uterine cervix cancer (UTC). Methods: The VEGF SiRNA chain 1, 2 and 3 were designed by web, based on software, and then synthesized. Then SiRNA were transfected into Hela cell line by lipofectamine 2000 reagent. Semi- quantity RT- PCR and Werstern blotting were used to analyzed the effect of the SiRNA transfection.Results: Significant down- regulation of the VEGF mRNA was found following transfection of the VEGF SiRNA chain 1 and 2. However, down regulation of the VEGF protein was slight and inconspicuous.Conclusions: The in vitro transient transfection of synthesized VEGF- SiRNAs on the UTC Hela cell line can down- regulate the expression of VEGFmRNA in the cells and the down- regulation rates of mRNA varies based on different SiRNA. As an important tool of the gene silencing, SiRNA is likely to be a new method of treating the UTC.
