Abstract:
Objective: To investigate the arsenic trioxide in the effect on the protein gene of drug resistance in the ls174 cell of human cancer of colon. Methods: Above all,to denifite noncytotoxic dose of As2O3, then treat ls174 cell of human cancer of colon with noncytotoxic dose of As2O3,the expression of MRP, GST- π and TOPO- Ⅱ was measured by biochemistry and RT- PCR. Results: The noncytotoxic dose of As2O3 (growth rate>95%) was 4.0μ /ml. In addition,a significant down regulation of MRP 、 GST- π 、 TOPO- ⅡmRNA was observed in the ls174 cell of human cancer of colon when 4.0μ /ml As2O3 was given. Conclusion: As2O3 is able to partly reverse the 5- FU resistance of the ls174 cell of human cancer of colon,which may be related to the variation of MRP 、 GST- π 、 TOPO- Ⅱ.
