三氧化二砷对人大肠癌ls174细胞凋亡及相关耐药蛋白基因表达的影响

To Investigate the Arsenic Trioxide In the Effect On Cells Apoptosis and the Protein Gene Of Drug Resistance In the Ls174 Cell of Human Cancer Of Colon

  • 摘要: 目的:探讨三氧化二砷(As2O3)对人大肠癌ls174细胞相关耐药蛋白基因表达的影响。方法:确定三氧化二砷(As2O3)对人大肠癌ls174细胞的非细胞毒剂量,分别采用生化方法及RT-PCR方法检测非细胞毒剂量下As2O3对人大肠癌ls174细胞MRP、GST-π、TOPO-Ⅱ表达水平的变化。结果:As2/sub>O3的非细胞毒性剂量(生长率>95%)为4.0μg/ml。以此为逆转耐药剂量可使人大肠癌ls174细胞,MRP、GST-π、TOPO-ⅡmRNA表达水平下调。结论:As2O3可部分逆转人大肠癌ls174细胞对5-氟尿嘧啶的耐药性,MRP、GST-π、TOPO-Ⅱ改变参与人大肠癌耐药机制。

     

    Abstract: Objective: To investigate the arsenic trioxide in the effect on the protein gene of drug resistance in the ls174 cell of human cancer of colon. Methods: Above all,to denifite noncytotoxic dose of As2O3, then treat ls174 cell of human cancer of colon with noncytotoxic dose of As2O3,the expression of MRP, GST- π and TOPO- Ⅱ was measured by biochemistry and RT- PCR. Results: The noncytotoxic dose of As2O3 (growth rate>95%) was 4.0μ /ml. In addition,a significant down regulation of MRP 、 GST- π 、 TOPO- ⅡmRNA was observed in the ls174 cell of human cancer of colon when 4.0μ /ml As2O3 was given. Conclusion: As2O3 is able to partly reverse the 5- FU resistance of the ls174 cell of human cancer of colon,which may be related to the variation of MRP 、 GST- π 、 TOPO- Ⅱ.
