Ca2+依赖的膜结合蛋白7在胃癌中的表达及临床意义

Expression and clinical significance of Ca2+-dependent membrane-binding protein 7 in gastric cancer

      目的  探讨Ca2+依赖的膜结合蛋7(CPNE7)在胃癌中的表达水平及其与肿瘤发生发展的关系。
      方法  首先通过生物信息学分析CPNE7在胃癌及癌旁中的表达差异及其与预后的关系。 收集2020年1月至2021年5月在兰州大学附属第二医院行手术治疗的102例胃癌患者的癌组织及其癌旁组织,制作成组织芯片(TMA),采用免疫组织化学染色法检测CPNE7的表达水平,Spearman相关分析CPNE7表达与胃癌患者临床病理特征的关系。采用RT-PCR和Western blot检测CPNE7在胃癌细胞(MKN45/AGS/HGC-27)和人正常胃黏膜细胞(GES-1)中mRNA和蛋白的表达水平。
      结果  CPNE7蛋白及其mRNA水平在胃癌中高于癌旁组织(P<0.05),CPNE7的表达与患者的临床分期(P<0.001)、T分期(P=0.012)、N分期(P=0.01)以及有无淋巴结浸润(P=0.005)和Lauren's分型(P=0.007)显著相关,与年龄(P=0.37)、性别(P=0.121)、分化程度(P=0.15)、有无转移(P=0.835)、血管浸润(P=0.275)、神经浸润无相关(P=0.814)。
      结论  CPNE7在胃癌组织和细胞中呈高表达,其表达与临床分期、T分期、N分期、淋巴结浸润和Lauren's分型相关,提示CPNE7可能参与胃癌的发生发展,有望成为胃癌的诊断标志物。

     

    Abstract:
      Objective  To investigate the expression level of Ca2+-dependent membrane-binding protein 7 (CPNE7) in gastric cancer and its relationship with tumor development.
      Methods  First, the expression difference of CPNE7 in gastric cancer and paracancerous tissues and its relationship with prognosis were analyzed using bioinformatics approach. The tumor tissues and adjacent tissues of 102 patients with gastric cancer who underwent surgical treatment at The Second Affiliated Hospital of Lanzhou University between January 2020 and May 2021 were collected and used to generate tissue chips (TMA). Immunohistochemical staining was used to detect the expression level of CPNE7. Spearman correlation analysis was performed for CPNE7 expression and clinicopathologic features of patients with gastric cancer. mRNA and protein expression levels of CPNE7 in gastric cancer cells (MKN45/AGS/HGC-27) and human normal gastric mucosa cells (GES-1) were determined by RT-PCR and Western blot.
      Results  CPNE7 protein and its mRNA levels in gastric cancer were higher than those in adjacent tissues (P<0.05). The expression of CPNE7 was significantly correlated with the clinical stage (P<0.001), T stage (P=0.012), N stage (P=0.01), lymph node infiltration (P=0.005), and Lauren's classification (P=0.007). There was no correlation with age (P=0.37), sex (P=0.121), differentiation degree (P=0.15), metastasis (P=0.835), vascular infiltration (P=0.275), or nerve infiltration (P=0.814).
      Conclusions  CPNE7 is highly expressed in gastric cancer tissues and cells, and its expression correlates with the clinical stage, T stage, N stage, lymph node invasion, and Lauren's classification, suggesting that CPNE7 may be involved in the occurrence and development of gastric cancer and can be expected to be a diagnostic marker for gastric cancer.
