Abstract:
Objective This study aimed to investigate the effect of the ectogenesis expression of miRNA-375 on cell proliferation and apoptosis in the human pancreatic cancer cell line Panc-1.
Methods After transfection of the miRNA-375 vector, the expression levels of miRNA-375 in the Panc-1 cells were determined by quantitative real-time polymerase chain reaction.Cell counting was performed to evaluate the effects of miRNA-375 expression on cell proliferation.Flow cytometry was used to analyze apoptosis and the cell cycle.
Results The expression levels of miRNA-375 were highly induced after miRNA-375 transfection.miRNA-375 restoration also inhibited cell proliferation, promoted cell apoptosis, and induced cell cycle arrest at G0-G1.
Conclusion The abnormal expression of miRNA-375 in the pancreatic cancer cell line Panc-1 inhibits the canceration of cells by interfering with cellular biological behaviors, such as proliferation and apoptosis.miRNA-375 may act as a potential tumor suppressor in the pancreatic cancer cell Panc-1.
