Effect of Twist1 on E-cadherin expression and subcellular location of human breast carcinoma cell line

      目的   探讨转录调控因子Twist1对人乳腺癌MCF-7细胞E-cadherin表达的影响。
      方法   上调人乳腺癌MCF-7细胞中Twist1的表达, 利用RT-PCR, Western blot, 细胞免疫荧光检测分析上调Twist1对E-cadherin表达的影响; 划痕实验分析对细胞侵袭运动能力的影响; 三维培养观察Twist1对MCF-7细胞成血管能力的影响。
      结果   MCF-7细胞过表达Twist1抑制细胞E-cadherin的表达, 并且导致其亚细胞定位异常, 细胞迁移运动能力增强, 细胞成血管能力增强。
      结论   Twist1能抑制MCF-7细胞E-cadherin的表达, 并增强其侵袭迁移及血管生成能力。

     

    Abstract:
      Objective   This study aims to investigate the effect of Twist1 on human breast carcinoma E-cadherin.
      Methods   pcDNA-Twist1 was transfected into human breast carcinoma cell line MCF-7.The expression of E-cadherin was detected via real-time polymerase chain reaction and immunofluorescence staining.Wound-healing assay was performed to examine the migration ability of cells, and the 3D culture was used to observe vasculogenic mimicry(VM) formation.
      Results   MCF-7 cells overexpressing Twist1 resulted in the suppression of E-cadherin expression and abnormal subcellular location.Migration ability and VM formation were also enhanced.
      Conclusion   Twist1 prevented E-cadherin expression as well as promoted migration ability and VM formation in human breast carcinoma cell line.
