Abstract:
Objective:To explore the effect of the transcription and nuclear factor (erythroid-derived 2)-like2 (Nrf 2) on (-) -epigallocat -echin- 3-gallate (EGCG)-inducing apoptosis in the esophageal cancer cell ECA- 109 . Methods:The MTT method is used to calculate the median lethal dose (IC50) in the blank control (control), negative control (si-NC), and ECA- 109 cells that are transfected with a si-RAN against Nrf2 (si-RNA). Flow cytometry is applied to test the effect of EGCG on apoptosis in three kinds of cells (control, si-NC, and si-RNA). Western blot is performed to test the effect of EGCG on the protein expression of Nrf 2 and cleaved Caspase- 3 in three kinds of cells (control, si-NC, and si-RNA). Results: In ECA- 109 cells, EGCG can induce the expression of Nrf2, and the deletion of Nrf2 signifi cantly reduced the IC 50 of EGCG, improved the apoptosis, and increased the expression of cleaved-Caspase-3. Conclusion: The Nrf2 in -duced by EGCG impaired the EGCG-inducing apoptosis in esophageal cancer cells.
