肾癌中EphA2/EphrinA1和E-cadherin的表达及其意义

Expression and Significance of EphrinA1, Ephrin Receptor A2 and E-cadherin in Human Renal Cell Carcinoma

  • 摘要: 目的: 探讨EphA2/EphrinA1和E-cadherin在肾癌组织中的表达及与肾癌临床病理特征的关系。 方法: 利用免疫组织化学法检测68例肾癌组织、24例正常肾组织中EphA2、EphrinA1和E-cadherin的表达;并采用流式细胞术对上述三种蛋白进行定量分析,采用SPSS13.0进行统计学处理。 结果: 肾癌组织中EphA2、EphrinA1的表达明显高于正常肾组织(P<0.01);E-cadherin的表达明显低于正常肾组织(P<0.01)。在肾癌组织中,肿瘤高分级组EphA2、EphrinA1蛋白的表达水平(IHC、FCM)显著高于低分级组(P<0.01,P<0.05),而E-cadherin蛋白的表达(IHC)显著低于低分级组(P<0.01);肿瘤高分期组EphA2、EphrinA1蛋白的表达水平(IHC、FCM)显著高于低分期组(P<0.01,P<0.05),而E-cadherin蛋白的表达(IHC)显著低于低分期组(P<0.01),淋巴结转移组EphA2、EphrinA1蛋白的表达(IHC、FCM)显著高于无淋巴结转移组(P<0.05),而E-cadherin蛋白的表达(IHC)显著低于无淋巴结转移组(P<0.05)。免疫组织化学显示EphA2与E-cadherin蛋白的表达呈显著负相关(r=-0.483,P<0.01),EphrinA1与E-cadherin蛋白的表达呈显著负相关(r=-0.414,P<0.01),EphA2与EphrinA1蛋白的表达呈显著正相关(r=0.906,P<0.01)。 结论: EphA2/EphrinA1与E-cadherin蛋白表达异常可能共同参与了肾癌的发生、发展与转移;联合检测三种蛋白对于评价肾癌的恶性程度、判断其转移潜能具有一定的参考价值。

     

    Abstract: Objective :To investigate the expression of EphrinA1,Ephrin receptor EphA2 and E-cadherin in renal cellcarcinoma and to explore its relevance to the clinicopathologic features. Methods :The expression of Ephri-nA1,EphA2 and E-cadherin was detected by immunohistochemistry (IHC) in tissues from 68 cases of renalcell carcinoma and 24 normal kidney tissue samples.Flow cytometry (FCM) was used to quantitate the ex-pression of EphrinA1,EphA2 and E-cadherin. Results :Compared with normal kidney tissues,renal cell carci-noma tissues had higher expression of EphrinA1and EphA2 (P<0.01) and lower expression of E-cadherin (P<0.01).Compared with carcinoma tissues of lower grade and less advanced stage or with no lymph nodemetastasis,tissues of higher grade and advanced stage or with lymph node metastasis had higher expressionof EphrinA1 and EphA2 (P<0.01) and lower expression of E-cadherin (P<0.05).The expression of these threeproteins was not associated with tumor diameter,age or gender (P>0.05).The expression of EphrinA1 orEphA2 was independently and inversely correlated with E-cadherin expression (r=-0.483,P<0.01 for EphA2,r=-0.414,P<0.01 for EphrinA1).A positive correlation was found between EphA2 expression and EphrinA1 ex-pression (r=0.906,P<0.01). Conclusion :EphrinA1,EphA2 and E-cadherin may be involved in the occurrence,development and metastasis of renal cell carcinoma.Simultaneous detection of the three proteins might behelpful for evaluating the malignance and predicting the metastatic potential of renal cell carcinoma.
